PyocyaninPromotesExtracellularDNAReleasein Pseudomonasaeruginosa TheerthankarDas*,MikeManefield CentreforMarineBioInnovation(CMB),SchoolofBiotechnologyandBiomolecularSciences(BABS),UniversityofNewSouthWales(UNSW),Sydney,Australia Abstract Bacterialadhesionandbiofilmformationarebothdependentontheproductionofextracellularpolymericsubstances(EPS) mainlycomposedofpolysaccharides,proteins,lipids,andextracellularDNA(eDNA).eDNApromotesbiofilmestablishment inawiderangeofbacterialspecies.InPseudomonasaeruginosaeDNAismajorcomponentofbiofilmsandisessentialfor biofilmformationandstability.InthisstudywereportthatproductionofpyocyanininP.aeruginosaPAO1andPA14batch culturesisresponsibleforpromotionofeDNArelease.AphzSHmutantofP.aeruginosaPAO1thatoverproducespyocyanin displayedenhancedhydrogenperoxide(H2O2)generation,celllysis,andeDNAreleaseincomparisontoitswildtypestrain. A DphzA-G mutant of P. aeruginosa PA14 deficient in pyocyanin production generated negligible amounts of H2O2 and releasedlesseDNAincomparisontoitswildtypecounterpart.ExogenousadditionofpyocyaninorincubationwithH2O2 wasalsoshowntopromoteeDNAreleaseinlowpyocyaninproducing(PAO1)andpyocynaindeficient(PA14)strains.Based onthesedataandrecentfindingsinthebiofilmliterature,weproposethattheimpactofpyocyaninonbiofilmformationin P.aeruginosaoccursviaeDNAreleasethroughH2O2mediatedcelllysis.
