RapidInternalizationoftheOncogenicK+Channel KV10.1 TobiasKohl1¤,EvaLo¨rinczi1,LuisA.Pardo1*,WalterStu¨hmer1,2* 1Max-Planck-Institute of Experimental Medicine, Department of Molecular Biology of Neuronal Signals, Go¨ttingen, Germany, 2DFG Research Center for Molecular PhysiologyoftheBrain(CMPB),Go¨ttingen,Germany Abstract KV10.1 is a mammalian brain voltage-gated potassium channel whose ectopic expression outside of the brain has been provenrelevantfortumorbiology.PromotionofcancercellproliferationbyKV10.1dependslargelyonionflow,butsome oncogenicpropertiesremainintheabsenceofionpermeation.Additionally,KV10.1surfacepopulationsaresmallcompared tolargeintracellularpools.Controlofproteinturnoverwithincellsiskeytobothcellularplasticityandhomeostasis,and thereforewesetouttoanalyzehowendocytictraffickingparticipatesincontrollingKV10.1intracellulardistributionandlife cycle.TofollowplasmamembraneKV10.1selectively,wegeneratedamodifiedchannelofdisplayinganextracellularaffinity tagforsurfacelabelingbya-bungarotoxin.ThismodificationonlyminimallyaffectedKV10.1electrophysiologicalproperties. Using a combination of microscopy and biochemistry techniques, we show that KV10.1 is constitutively internalized involvingatleasttwodistinctpathwaysofendocytosisandmainlysortedtolysosomes.Thisoccursatarelativelyfastrate. Simultaneously,recyclingseemstocontributetomaintainbasalKV10.1surfacelevels.BriefKV10.1surfacehalf-lifeandrapid lysosomaltargetingisarelevantfactortobetakenintoaccountforpotentialdrugdeliveryandtargetingstrategiesdirected againstKV10.1ontumorcells.
