Olofssonetal.BMCPlantBiology2011,11:45 /1471-2229/11/45 RESEARCH ARTICLE OpenAccess Relativeexpressionofgenesofterpene metabolismindifferenttissuesofArtemisia annuaL LindaOlofsson,AlexanderEngstr?m,AnneliLundgren,PeterEBrodelius* Abstract Background:Recently,ArtemisiaannuaL.(annualorsweetwormwood)hasreceivedincreasingattentiondueto thefactthattheplantproducesthesesquiterpenoidendoperoxideartemisinin,whichtodayiswidelyusedfor treatmentofmalaria.Theplantproducesrelativelysmallamountsofartemisininandaworldwideshortageofthe drughasledtointenseresearchinordertoincreasetheyieldofartemisinin.Inordertoimproveour understandingofterpenemetabolismintheplantandtoevaluatethecompetitionforprecursors,whichmay influencetheyieldofartemisinin,wehaveusedqPCRtoestimatetheexpressionof14genesofterpene metabolismindifferenttissues. Results:Thefourgenesoftheartemisininbiosyntheticpathway(amorpha-4,11-dienesynthase,amorphadiene-12- hydroxylase,artemisinicaldehydeΔ11(13)reductaseandaldehydedehydrogenase1)showedremarkablyhigher expression(between~40-to~500-fold)inflowerbudsandyoungleavescomparedtoothertissues(oldleaves, stems,roots,hairyrootcultures).Further,dihydroartemisinicaldehydereductaseshowedaveryhighexpression onlyinhairyrootcultures.GermacreneAandcaryophyllenesynthaseweremostlyexpressedinyoungleavesand flowerbudswhileepi-cedrolsynthasewashighlyexpressedinoldleaves.3-Hydroxy-3-methyl-glutarylcoenzymeA reductaseexhibitedlowerexpressioninoldleavescomparedtoothertissues.Farnesyldiphosphatesynthase, squalenesynthase,and1-deoxy-D-xylulose-5-phosphatereductoisomeraseshowedonlymodestvariationin expressioninthedifferenttissues,whileexpressionof1-deoxy-D-xylulose-5-phosphatesynthasewas7-8-fold higherinflowerbudsandyoungleavescomparedtooldleaves. Conclusions:Fourgenesofartemisininbiosynthesiswerehighlyexpressedinflowerbudsandyoungleaves (tissuesshowingahighdensityofglandulartrichomes).Theexpressionofdihydroartemisinicaldehydereductase hasbeensuggestedtohaveanegativeeffectonartemisininproduct
