ResolutionofUltramicroscopyandFieldofViewAnalysis UlrichLeischner1,2*,WalterZieglga¨nsberger1,Hans-UlrichDodt1,3 1MaxPlanckInstituteofPsychiatry, Munich,Germany, 2TUMu¨nchen,Institute forNeuroscience,Munich,Germany, 3Department ofBioelectronics,InstituteofSolid StateElectronics,TUVienna,Vienna,Austria Abstract In a recent publication we described a microscopical technique called Ultramicroscopy, combined with a histological procedurethatmakesbiologicalsamplestransparent.Withthiscombinationwecangatherthree-dimensionalimagedata oflargebiologicalsamples.Herewepresentthetheoreticalanalysisofthez-resolution.Byanalyzingthecross-sectionofthe illuminating sheet of light we derive resolution values according to the Rayleigh-criterion. Next we investigate the resolutionadjacenttothefocalpointoftheilluminationbeam,analyzethroughoutwhatextendtheilluminationbeamisof acceptablesharpnessandinvestigatetheresolutionimprovementscausedbytheobjectivelens.Finallyweconcludewitha usefulruleforthesamplingrates.ThesefindingsareofpracticalimportanceforresearchersworkingwithUltramicroscopy todecideonadequatesamplingrates.Theyarealsonecessarytomodifydeconvolutiontechniquestogainfurtherimage improvements.
