RepressionofFLOWERINGLOCUSTChromatinby FunctionallyRedundantHistoneH3Lysine4 DemethylasesinArabidopsis Ju-HeeJeong1,2.,Hae-RyongSong1,2.,Jong-HyunKo1,2,Young-MinJeong1,2,4,YoungEunKwon1 ,Jae HongSeol1,RichardM.Amasino2,4,BoslNoh2,3*,Yoo-SunNoh1,2 * 1School of Biological Sciences,Seoul National University, Seoul, Korea, 2GlobalResearch Laboratory for FloralRegulatory Signaling,Seoul National University, Seoul, Korea, 3Environmental Biotechnology National Core Research Center, Gyeongsang National University, Jinju, Korea, 4Department of Biochemistry, University of Wisconsin,Madison,Wisconsin,UnitedStatesofAmerica Abstract FLOWERINGLOCUST(FT)playsakeyroleasamobilefloralinductionsignalthatinitiatesthefloraltransition.Therefore, precisecontrolofFTexpressioniscriticalforthereproductivesuccessoffloweringplants.Coexistenceofbivalenthistone H3 lysine 27 trimethylation (H3K27me3) and H3K4me3 marks at the FT locus and the role of H3K27me3 as a strong FT repressionmechanisminArabidopsishavebeenreported.However,theroleofanactivemark,H3K4me3,inFTregulation hasnotbeenaddressed,norhavethecomponentsaffectingthismarkbeenidentified.MutationsinArabidopsisthaliana Jumonji4(AtJmj4)andEARLYFLOWERING6(ELF6),twoArabidopsisgenesencodingJumonji(Jmj)familyproteins,causedFT- dependent,additiveearlyfloweringcorrelatedwithincreasedexpressionofFTmRNAandincreasedH3K4me3levelswithin FTchromatin.PurifiedrecombinantAtJmj4proteinpossessesspecificdemethylaseactivityformono-,di-,andtrimethylated H3K4.TaggedAtJmj4andELF6proteinsassociatedirectly withthe FTtranscriptioninitiationregion, aregionwherethe H3K4me3levelswereincreasedmostsignificantlyinthemutants.Thus,ourstudydemonstratestherolesofAtJmj4and ELF6asH3K4demethylasesdirectlyrepressingFTchromatinandpreventingprecociousfloweringinArabidopsis.
