RoleofCajalBodiesandNucleolusintheMaturationof theU1snRNPinArabidopsis ZdravkoJ.Lorkovic′*,AndreaBarta MaxF.PerutzLaboratories,DepartmentofMedicalBiochemistry,MedicalUniversityofVienna,Vienna,Austria Abstract Background:ThebiogenesisofspliceosomalsnRNPstakesplaceinboththecytoplasmwhereSmcoreproteinsareadded andsnRNAsaremodifiedatthe59and39terminiandinthenucleuswheresnRNP-specificproteinsassociate.U1snRNP consists of U1 snRNA, seven Sm proteins and three snRNP-specific proteins, U1-70K, U1A, and U1C. It has been shown previouslythatafterimporttothenucleusU2andU4/U6snRNP-specificproteinsfirstappearinCajalbodies(CB)andthen insplicingspeckles.Inaddition,incellsgrownundernormalconditionsU2,U4,U5,andU6snRNAs/snRNPsareabundantin CBs. Therefore, it has been proposed that the final assembly of these spliceosomal snRNPs takes place in this nuclear compartment.Incontrast,U1snRNAinbothanimalandplantcellshasrarelybeenfoundinthisnuclearcompartment. Methodology/PrincipalFindings:Here,weanalysedthesubnucleardistributionofArabidopsisU1snRNP-specificproteins fused to GFP or mRFP in transiently transformed Arabidopsis protoplasts. Irrespective of the tag used, U1-70K was exclusivelyfoundinthenucleus,whereasU1AandU1Cwereequallydistributedbetweenthenucleusandthecytoplasm.In thenucleusallthreeproteinslocalisedtoCBsandnucleolialthoughtodifferentextent.Interestingly,wealsofoundthatthe appearanceofthethreeproteinsinnuclearspecklesdiffersignificantly.U1-70KwasmostlyfoundinspeckleswhereasU1A andU1Cin,90%ofcellsshoweddiffusenucleoplasmicincombinationwithCBsandnucleolarlocalisation. Conclusions/Significance:OurdataindicatethatCBsandnucleolusareinvolvedinthematurationofU1snRNP.Differences innuclearaccumulationanddistributionbetweenU1-70KandU1AandU1CproteinsmayindicatethateitherU1-70Kor U1AandU1Cassociatewith,oris/areinvolved,inothernuclearprocessesapartfrompre-mRNAsplicing.
