Erdeietal.BiotechnologyforBiofuels2012,5:12 /content/5/1/12(12March2012) Erdeietal.BiotechnologyforBiofuels2012,5:12 /content/5/1/12 RESEARCH OpenAccess Separatehydrolysisandco-fermentationfor improvedxyloseutilizationinintegratedethanol productionfromwheatmealandwheatstraw BorbálaErdei1*,BalázsFrankó2,MatsGalbe1andGuidoZacchi1 Abstract Background:Thecommercializationofsecond-generationbioethanolhasnotbeenrealizedduetoseveralfactors, includingpoorbiomassutilizationandhighproductioncost.Itisgenerallyacceptedthatthemostimportant parametersinreducingtheproductioncostaretheethanolyieldandtheethanolconcentrationinthe fermentationbroth.Agriculturalresiduescontainlargeamountsofhemicellulose,andtheutilizationofxyloseis thusaplausiblewaytoimprovetheconcentrationandyieldofethanolduringfermentation.Mostnaturally occurringethanol-fermentingmicroorganismsdonotutilizexylose,butageneticallymodifiedyeaststrain, TMB3400,hastheabilitytoco-fermentglucoseandxylose.However,thexyloseuptakerateisonlyenhanced whentheglucoseconcentrationislow. Results:Separatehydrolysisandco-fermentationofsteam-pretreatedwheatstraw(SPWS)combinedwithwheat- starchhydrolysatefeedwasperformedintwoseparateprocesses.Theaverageyieldofethanolandthexylose consumptionreached86%and69%,respectively,whenthehydrolysateoftheenzymaticallyhydrolyzed(18.5% WIS)unwashedSPWSsolidfractionandwheat-starchhydrolysatewerefedtothefermentorafter1hof fermentationoftheSPWSliquidfraction.Intheotherconfiguration,fermentationoftheSPWShydrolysate(7.0% WIS),resultedinanaverageethanolyieldof93%fromfermentationbasedonglucoseandxyloseandcomplete xyloseconsumptionwhenwheat-starchhydrolysatewasincludedinthefeed.Increasedinitialcelldensityinthe fermentation(from5to20g/L)didnotincreasetheethanolyield,butimprovedandacceleratedxylose consumptioninbothcases. Conclusions:Higherethanolyieldhasbeenachievedinco-ferment
