Results:Here,weevaluatedchangesingeneexpressionoverthecourseofseeddevelopmentusingIllumina (formerlySolexa)RNA-sequencing.Tissuesat15daysafterflowering(DAF)servedasthecontrol,andatotalof 11592,16594,and16255differentiallyexpressedunigeneswereidentifiedat35,55,and65DAF,respectively.Gene Ontologyanalysesdetected113co-expressedunigenesassociatedwithlipidbiosynthesis.Ofthese,15showed significantchangesinexpressionlevels(log2foldvalues≥1)duringseeddevelopment.Pathwayanalysisrevealed 24co-expressedtranscriptsinvolvedinlipidbiosynthesisandfattyacidbiosynthesispathways.Weselected12 differentiallyexpressedgenesandanalyzedtheirexpressionsusingqRT-PCR.Theresultswereconsistentwiththose obtainedfromSolexasequencing. Conclusion:Theseresultsprovideacomprehensivemolecularbiologybackgroundforresearchonsoybeanseed development,particularlywithrespecttotheprocessofoilaccumulation.Allofthegenesidentifiedinourresearch havesignificanceforbreedingsoybeanswithincreasedoilcontents.
