Methodology:WeinvestigatedECresponsetolong-terminflammatorycuesunderphysiologicallyrelevantshearconditions viaE-selectinexpressionwheremonolayersofhumanumbilicalveinECsweresimultaneouslyexposedtolaminarfluidshear andinterleukin-1?(shear-cytokineactivation)inaparallelplateflowchamber. ResultsandConclusion:Na?¨veECsexposedtoshear-cytokineactivationdisplaysignificantlyhigherE-selectinexpression for up to 24hr relative to ECs activated in static (static-cytokine). Peak E-selectin expression occurred after 8–12hr of continuousshear-cytokineactivationcontrarytothecommonlyobserved4–6hrpeakexpressioninECsexposedtostatic- cytokineactivation.CellswithsomehistoryofhighshearconditioningexhibitedeitherhighormutedE-selectinexpression dependingonthedurationsoftheshearpre-conditioningandtheensuingshear-cytokineactivation.Overall,thepresented datasuggestthatahighlaminarshearenhancesacuteECresponsetointerleukin-1?inna?¨veorshear-conditionedECsas maybefoundinthepathologicalsettingofischemia/reperfusioninjurywhileconferringrapidE-selectindownregulationto protectagainstchronicinflammation.
