MethodsandFindings:Inthepresentstudy,weshowedthatmaturationofoligodendrocyteswasattenuatedbysodium orthovanadate (a comprehensive inhibitor of tyrosine phosphatases) and PTPi IV (a specific inhibitor of SHP-2). It is also foundthatSHP-2waspersistentlyexpressedduringmaturationprocessofOPCs.Down-regulationofendogenousSHP-2led toimpairmentofoligodendrocytesmaturationandthiseffectwastriiodo-L-thyronine(T3)dependent.Furthermore,over- expressionofSHP-2wasshowntopromotematurationofoligodendrocytes.Finally,ithasbeenidentifiedthatSHP-2was involvedinactivationofAktandextracellular-regulatedkinases1and2(ERK1/2)inducedbyT3inoligodendrocytes.
