SignificantExpressionLevelsofTransgenicPPP1CC2in TestisandSpermAreRequiredtoOvercometheMale InfertilityPhenotypeofPpp1ccNullMice NilamSinha1*¤,StephenPilder2,SrinivasanVijayaraghavan1* 1DepartmentofBiologicalSciences,KentStateUniversity,Kent,Ohio,UnitedStatesofAmerica,2DepartmentofAnatomyandCellBiology,TempleUniversitySchoolof Medicine,Philadelphia,Pennsylvania,UnitedStatesofAmerica Abstract PPP1CC2,oneoffourisoformsoftheser/thrproteinphosphatasePP1,isamammalian-specificsplicevariantofthePpp1cc gene,andtheonlyisoformwhoseexpressionisconfinedalmostcompletelytospermatogeniccells.Additionally,PPP1CC2 isthesoleisoformfoundinmammalianspermatozoa.AlthoughPPP1CC1,theotherPpp1ccproduct,isexpressedinmany tissuesincludingtestis,theonlyphenotyperesultingfromdeletionofPpp1ccgeneismaleinfertility.Todeterminewhichof the products of Ppp1cc is essential for male fertility, we created two PPP1CC2 transgenes, eTg-G2 and pTg-G2, where Ppp1cc2 expression was driven by the putative endogenous promoter of Ppp1cc or by the testis specific human Pgk2 promoter,respectively.Ourresultsdemonstratethatthe2.6-kbgenomicregiondirectlyupstreamofthePpp1ccstructural genecandriveexpressionofPpp1cc2,andrecapitulatethewild-typetissuespecificityofPPP1CC2intransgenicmice.More importantly, we show that expression of PPP1CC2 alone, via either promoter, is able not only to restore normal spermatogenesis,butthefertilityofPpp1ccnullmiceaswell,providedthattransgenicPPP1CC2expressionintestisreaches atleastalowerthresholdlevelequivalenttoapproximately50%ofitsexpressionbyaPpp1cc+/2male.Weconcludethat theendogenousPpp1ccpromoternormallyfunctionsinthetestistomaintainasufficientlevelofPPP1CC2expressionfor normalspermatogenesistooccur,andthatproductionofspermatozoacapableoffertilizationinvivocantakeplaceinthe completeabsenceofPPP1CC1expression.
