Teohetal.BMCPlantBiology2013,13:19 /1471-2229/13/19 RESEARCH ARTICLE OpenAccess Transcriptomeanalysisofembryomaturationin maize KeatThomasTeoh1,DeborahVicunaRequesens1,ShivakumarPDevaiah1,DanielJohnson1,XiuzhenHuang1, JohnAHoward2andElizabethEHood1* Abstract Background:Maizeisoneofthemostimportantcropsintheworld.Withtheexponentiallyincreasingpopulation andtheneedforeverincreasedfoodandfeedproduction,anincreasedyieldofmaizegrain(aswellasrice,wheat andothergrains)willbecritical.Maizegraindevelopmentisunderstoodfromtheperspectiveofmorphology, hormoneresponses,andstoragereserveaccumulation.Thisincludesvariousstudiesongeneexpressionduring embryodevelopmentandmaturationbutaglobalstudyofgeneexpressionoftheembryohasnotbeenpossible untilrecently.Transcriptomeanalysisisapowerfulnewtoolthatcanbeusedtounderstandthegeneticbasisof embryomaturation. Results:Weundertookatranscriptomicanalysisofnormalmaturingembryosat15,21and27daysafter pollination(DAP),ofoneelitemaizegermplasmlinethatwasutilizedincrossestotransgenicplants.Morethan 19,000geneswereanalyzedbythismethodandthechallengewastoselectsubsetsofgenesthatarevitally importanttoembryodevelopmentandmaturationfortheinitialanalysis.Wedescribethechangesinexpression forgenesrelatingtoprimarymetabolicpathways,DNAsynthesis,lateembryogenesisproteinsandembryostorage proteins,shownthroughtranscriptomeanalysisandconfirmedlevelsoftranscriptionforsomegenesinthe transcriptomeusingqRT-PCR. Conclusions:Numerousgenesinvolvedinembryomaturationhavebeenidentified,manyofwhichshowchanges inexpressionlevelduringtheprogressionfrom15to27DAP.Anexpectedarrayofgenesinvolvedinprimary metabolismwasidentified.Moreover,morethan30%oftranscriptsrepresentedun-annotatedgenes,leavingmany functionstobediscovered.Ofparticularinterestarethestorageproteingenes,globulin-1,globulin-2andan unidentifiedcupinfamilygene.Whenexpressingforeignproteinsinmaize,theglobulin-1promoterismostoften used,butthiscupinfamilygenehasmuchhigherexpressionandmaybeabettercandidateforforeigng
