Abstract
Background:Althoughthegeneexpressionprofileofseveraltissuesinhumansandinrodentanimalmodelshas
beenexplored,analysisofthecompletechoroidplexus(CP)transcriptomeisstilllacking.Abettercharacterization
oftheCPtranscriptomecanprovidekeyinsightsintoitsfunctionsasoneofthebarriersthatseparatethebrain
fromtheperipheryandintheproductionofcerebrospinalfluid.
Methods:ThisworkextendsfurtherwhatisknownaboutthemouseCPtranscriptomethroughamicroarray
analysisofCPtissuefromnormalmiceunderphysiologicalconditions.
Results:Wefoundthatthegenesmosthighlyexpressedarethoseimplicatedinenergymetabolism(oxidative
phosphorylation,glycolysis/gluconeogenesis)andinribosomalfunction,whichisinagreementwiththesecretory
natureoftheCP.Ontheotherhand,genesencodingforimmunemediatorsareamongthosewithlower
expressioninbasalconditions.Inaddition,wefoundgenesknowntoberelevantduringbraindevelopment,and
notpreviouslyidentifiedtobeexpressedintheCP,includingthoseencodingforvariousaxonalguidanceand
angiogenesismoleculesandforgrowthfactors.Someoftheseareknowntoinfluencetheneuralstemcellniche
inthesubventricularzone,highlightingtheinvolvementoftheCPasalikelymodulatorofneurogenesis.
Interestingly,ourobservationsconfirmthattheCPtranscriptomeisunique,displayinglowhomologywiththatof
othertissues.Ofnote,wedescribeherethattheclosestsimilarityiswiththetranscriptomeoftheendothelialcells
oftheblood-brainbarrier.
Conclusions:Basedonthedatapresentedhere,itwillnowbepossibletofurtherexplorethefunctionof
particularproteinsoftheCPsecretomeinhealthandindisease.
