Results:Aphylogeneticanalysishasshownthatthetwoputativeaquaporinsbelongtodifferentfunctional families:NIPsandXIPs.Transcriptomicexperimentshaveshowntheindependenceoftheirexpressionfromtheir nutritionalstatusbutalsoaclosecorrelationwithmycorrhizalandrhizobialinteraction.Furthertranscript quantificationhasrevealedagoodcorrelationbetweentheexpressionofoneofthem,LjNIP1,andLjPT4 ,the phosphatetransporterwhichisconsideredamarkergeneformycorrhizalfunctionality.Byusinglaser microdissection,wehavedemonstratedthatoneofthetwogenes,LjNIP1,isexpressedexclusivelyin arbuscule-containingcells.LjNIP1,inagreementwithitsputativeroleasanaquaporin,iscapableoftransferring waterwhenexpressedinyeastprotoplasts.ConfocalanalysishavedemonstratedthateGFP-LjNIP1,underits endogenouspromoter,accumulatesintheinnermembranesystemofarbusculatedcells. Conclusions:Overall,theresultshaveshowndifferentfunctionalityandexpressionspecificityoftwo mycorrhiza-inducibleaquaporinsinL.japonicus.Oneofthem,LjNIP1canbeconsideredanovelmolecularmarker ofmycorrhizalstatusatdifferentdevelopmentalstagesofthearbuscule.Atthesametime,LjXIP1resultstobethe firstXIPfamilyaquaporintobetranscriptionallyregulatedduringsymbiosis.
