Results:Synechocystissp.PCC6803mutantstrainscontainingeitheroverexpressionordeletionoftheslr1609gene, whichencodesanacyl-ACPsynthetase(AAS),havebeenconstructed.Thecompletesegregationanddeletioninall mutantstrainswasconfirmedbyPCRanalysis.Blockingfattyacidactivationbydeletingslr1609geneinwild-type Synechocystissp.PCC6803ledtoadoublingoftheamountoffreefattyacidsandadecreaseofalkaneproduction byupto90percent.Overexpressionofslr1609geneinthewild-typeSynechocystissp.PCC6803hadnoeffecton theproductionofeitherfreefattyacidsoralkanes.Overexpressionordeletionofslr1609geneintheSynechocystis sp.PCC6803mutantstrainwiththecapabilityofmakingfattyalcoholsbygeneticallyintroducingfattyacyl-CoA reductaserespectivelyenhancedorreducedfattyalcoholproductionby60percent. Conclusions:Fattyacidactivationfunctionalizedbytheslr1609geneismetabolicallycrucialforbiosynthesisof fattyacidderivativesinSynechocystissp.PCC6803.Itisnecessarybutnotsufficientforefficientproductionof alkanes.Fattyalcoholproductioncanbesignificantlyimprovedbytheoverexpressionofslr1609gene.
