安徽农业大学学报, 2017, 44(2): 203-207 Journal of Anhui Agricultural University [DOI] 10.13610/ki.1672-352x020 网络出版时间：2017/4/19 12:00:21 [URL] /kcms/detail/34.1162.S1200.040.html 部分鸡种Brd2 基因 SNP 和 INDEL 分析 吴 允，毕榆林，张 扬，郭晓敏，李志腾，万 方， * 朱鹏飞，徐 璐 ，常国斌 ，徐 琪，陈国宏 (扬州大学动物科学与技术学院，扬州 225009) 摘 要：Brd2 （bromodomain containing protein 2 ）基因位于MHC （major histocompatibility complex ）基因家族 中，探究不同鸡种Brd2 基因的SNP 和Indel 位点具有重要意义。试验选取我国26 个优良的地方鸡种，对Brd2 基 因进行目标捕获测序，以NCBI 中原鸡（Gallus gallus）的Brd2 基因序列为参照，筛选出Brd2 基因外显子和内含 子中的SNP 和Indel 以及氨基酸的改变位点，最后制作进化树。在26 个鸡种中，Brd2 基因的外显子筛选出25 个 SNP 位点，没有检测到Indel 位点；内含子筛选出30 个Indel 位点，没有筛选出SNP 位点；外显子中氨基酸仅有4 个发生错义突变，其余均发生同义突变。从进化树看出，26 个鸡种分为 3 大类：安卡鸡、雪山草鸡、文昌鸡、北 京油鸡和萧山鸡类聚在一起；泰和乌骨鸡、SPF-来航鸡、河南斗鸡和广西黄鸡类聚在一起；剩余的鸡种类聚在一起， 但同源性相对较低。在我国优质鸡种中Brd2 基因具有一定的保守性，不同鸡种同源性较低。 关键词：鸡；Brd2 基因；SNP ；Indel ；进化树 中图分类号：S831.2 文献标识码：A 文章编号：1672352X (2017)02020305 SNP and INDEL analysis of Brd2 gene in indigenous chicken breeds WU Yun, BI Yulin, ZHANG Yang, GUO Xiaomin, LI Zhiteng, WAN Fang, ZHU Pengfei, XU Lu, CHANG Guobin, XU Qi, CHEN Guohong (College of Animal Science and Technology, Yangzhou University, Yangzhou 225009) Abstract: Inquiring the SNP and Indel loci of the Brd2 gene belonging to MHC family is of great signifi- cance.
