扬子鳄鞣制皮革和鳞片的DNA提取方法.PDFVIP

动物学报 　50 (2) :297 - 301 , 2004 A cta Zoologica S inica 　　　　　 　 扬子鳄鞣制皮革和鳞片的 D NA 提取方法 史　燕　吴孝兵 　晏　鹏　赵　哲 安徽师范大学生命科学学院 , 安徽芜湖 241000 摘 　要 　运用一种改进的提取方法 , 作者从鞣制皮革中成功地提取了总 DNA , 同时还对尾尖皮、鳞片、盐腌生 皮等皮质进行了 DNA 提取 ; 用 12S rRNA 基因扩增的通用引物、扬子鳄鉴别引物、微卫星引物及 RAPD 引物进 行 PCR 扩增 , 并对部分扩增结果进行测序 , 以检验提取效果。结果证明, 几种皮质标本都可提取出 DNA , 其中 尾尖皮和鳞片的提取效果较好 , 用四种引物都可扩增出明显亮带 ; 盐腌生皮和鞣制皮提取结果也很好 , 并且用 12S rRNA 通用引物、扬子鳄鉴别引物扩增的亮带较明显 , 可进行扬子鳄皮质用品等的分子鉴定及部分序列的扩 增和测序研究 [动物学报 50 (2) : 297 - 301 , 2004 ] 。 关键词 　扬子鳄 　皮革 　总 DNA 　PCR A method for D NA extraction from tanna ge and scales of Chinese alligator Alli gator sinensis SHI Yan , WU XiaoBing , YAN Peng , ZHAO Zhe College of Life Sciences , Anhui Normal University , Wuhu 　241000 , Anhui , China Abstract 　Total DNA from tannage , tail skin , scales and the skin treated with salt were successfully extracted with an improved method. To verify the result , four pair of primers , which were universal primers for 12S rRNA gene , identifi cation primers of Chinese alligator A lli gator sinensis meat , microsatellite primer and RAPD primer were used to do ampli fications. Some amplified fragments were sequenced. The results show that DNA of these samples can be extracted by this method. DNA of tail skin and scales can be amplified well with four primers. The skin treated with salt and tannage DNA can also be extracted and amplified with universal primers and identification primers of A lli gator sinensis meat.
