组比有显著的统计学差异(P0.05) ，细胞周期阻滞在G0/G1 期；对照组细胞的凋 亡率为 6.5 ±0.10% ，而 0.4 μmol/L5-Aza-CdR 处理组细胞的凋亡率为 11.8± 0.30% ，两者间有显著的统计学差异(P0.01) 。以上作用与药物浓度呈剂量依赖 性关系。 RT-PCR 分析显示5-Aza-CdR 处理前，乳腺癌细胞株MCF-7 无RASSF1A 表达，处理后的可检测出基因RASSF1A 的重新表达。 结论：5-氮杂2-脱氧胞苷可消除RASSF1A 启动子甲基化状态，使其重新表达 而抑制乳腺癌细胞株MCF-7 的生长，使细胞周期阻滞于G0/G1 期，并可促进其 凋亡。 关键词: 5-氮杂-2-脱氧胞苷；乳腺癌细胞；RASSF1A ；细胞周期；细胞凋亡； 4 Effect of 5-Aza-2-deoxycytidine on biological behaviors of human breast cancer cell line MCF-7 Abstract AIM: To explore the effect of methylation inhibitor, 5-Aza-2-deoxycytidine (5-Aza-CdR) on the growth,cell cycle and expression of RASSF1AmRNA of human breast cancer MCF-7 cells, in order to investigate the mechanism and to provide principle of its application in clinically treating breast cancer. METHODS: Human breast cancer MCF-7 cells were treated with 0.4, 1.6, 6.4, 25.6,and 102.4 μmol/L 5-Aza-CdR, respectively. Then the growth of the cells were observed by MTT assay.The cell cycle and apoptosis were analyzed by flow cytometry. The expression of the tumor suppressor gene RASSF1AmRNA was detected by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: After 5-Aza-CdR treatment, malignant degree and activeness of human breast cancer MCF-7 cells reducesed while response time and the concentration increased.Then the cells displayed early apoptosis and necrotized at a concentration of 102.4 μmol/L.
