忧虑康药对动物抑郁行为及血浆cAMP、cGMP影响及毒性试验探究 　 作者：史冰 陈晓阳 李晟 秦莉花 吴晓军 陈容 邹志 【摘要】 目的 观察忧虑康药（浸膏）对肾阴虚型抑郁模型大鼠行为学和血浆cAMP、cGMP的 影响 及毒性试验。 方法 采用灌胃甲状腺素合并慢性不可预见性应激制成肾阴虚抑郁动物模型后，随机分为空白对照组、模型组、六味地黄丸组、氟西汀组、忧虑康低、中、高剂量组，观察各组大鼠敞箱试验和液体消耗试验等行为学变化，采用放射免疫方法检测血浆cAMP、cGMP含量。并进行急性毒性试验， 计算 小鼠最大耐受量。结果 肾阴虚抑郁模型大鼠敞箱试验和液体消耗试验各项指标均显著下降，血浆cAMP含量显著升高；忧虑康药能部分改善大鼠的抑郁行为，六味地黄丸和忧虑康均能显著降低大鼠血浆cAMP含量。小鼠灌服忧虑康药1日最大耐受量为360g/kg，相当于人临床用量（1.2g/kg）的300倍。结论 呈量效关系的忧虑康药影响大鼠行为学和调整血浆cAMP、cGMP平衡，可能正是其滋阴解郁的作用机制之一；该药是安全范围较大的药物，提示用于临床是安全的。 毕业论文 【关键词】 忧虑康；肾阴虚；抑郁症；环腺苷酸；环鸟苷酸；急毒 毕业论文 The effects and toxicity experimental research of You Lv Kang on animal depression behavior and cAMP,cGMP in the plasma 【Abstract】 Objective To objective the effects of You Lv Kang on the cAMP、cGMP in the plasma of modeling rats following renal yin deficiency and on the behavior of these animals and toxicity test.Methods Rats were divided into seven groups randomly: control,model,Liu WEi Di Huang Wan,fiuoxetine, the low、middle and high-dose group of You Lv Kang.To establish the modeling rats following renal yin deficiency by exposing the animals to chronic stress depression and giving them by intragastric adminstration.To observe the behavior changs in the open-field test and fluid comsumption test. the plasma samples of experimental rats were taken for cAMP and cGMP determination by radioimmunoassay.To compute maximally tolerated dose of rats by acute toxicity test. Results All the indexes of the model rats in the open-field test and fluid comsumption test were significantly decreased, the plasma concentration of cAMP was significantly elevated;You Lv Kang can partly improve the behavior of the rats,the plasma of cAMP were significantly decreased in the Liu WEI Di Huang Wan and You Lv Kang groups.The maximally tolerated dose of mouse were 360g/kg every day,equivaent 300 times of clinic doses（1.2g/kg）.Conclusion One of the mechanism of increasing yin and improving depression was the effects of The You Lv Kang on the behavior changs of the rats
