热带作物学报 2014，35(10)：1969—1974 ChineseJournalofTropicalCrops 番茄S1ARF2基因表达模式及其 在果实发育中的功能分析 杨 枭，任振新 ，林冬波，先志强，李正国 妻重庆享市高校功能鑫基篙因蔫与簧调控囊新技术重点实墓验室 重王庆400030 摘 要 为 了分析 SlARF2基 因在番茄生长发育过程 中的功能．利用定量 PCR技术分析 SlARF2基 因的表达模 式，发现SlARF2基 因在番茄花芽中表达量最高，在幼果 中次之 ，说明SlARF2基 因可能在番茄花果发育中起着 重要作用。为进一步研究 SlARF2的功能 ，构建 S1ARF2基 因超表达 、抑制表达载体 ，农杆菌介导转化番茄成功 获得了相应转基 因植株 。通过与野生型番茄植株花 、果不 同时期 的表 型进行 比对分析 ，发现 SlARF2基 因的超 表达对花的形态学特征无明显影响 。但能够显著提高植株的结实率、降低果实大小和重量。说明 s|A／RF2基因 参与调控番茄花、果发育过程 。 关键词 番茄 ；SA／RF2；表达模式 ；果实发育 中图分类号 $641．2 文献标识码 A TheExpression Pattern ofSIARF2 in Tomato and the FunctiOn Analysisin Tomato FruitDevelopment YANG Xiao，REN Zhenxin，LIN Dongbo，XIAN Zhiqiang，LIZhengguo GeneticEngineeringResearchCenter,SchoolofLfieSciences，ChongqingUniversity／KeyLabofFu~twndGene andNewRegulationTechnologiesof ChongqingMunicipalEducationCommission,Chongqing400030,China Abstract To investigate thefunction ofSbtRF2 in tomato development，weanalyzed the expression pattern of S 足 by Real—time PCR．We ofund thatS／ARF2 washighly expressed in tomato bud and immature fruit． suggestingthatSlARF2playsan importantroleintomatoflowerand fruitdevelopment．Furthermore，we constructed SlARF2 overexpression and suppress expression vectors．By agrobacterium infection，we finally obtained the SlARF2 overexpression and suppress transgenetic tomatoes．Compared the transgenetic tomatoes to wild type tomato，we found thatSlARF2 had no affection in hte morphology offlower，however SlARF2 overexpression strongly enhanced the rape rate oftomato and decreased hte size oftomato furit．1’I1e study demonstrates that SlAR1；2 regulates hte developmentoftomato flowerand furit，and providesbasic reference forfurtherstudy of ．s F2 function． Key words Tomato；SlARF2；Expression pattern；Furitd
