JournalofFoodScienceandEngineering3(2013)616-624 NG DeterminationofAntioxidantandCytotoxicActivitiesof RedSeaweed，(Gracilariamanilaensis)againstDifferent CancerCellLines NorSalmiAbdullah，ShamsulM uhamad，Ibrahim CheOmar andHasmahAbdullah2 ，．FacultyAgro—basedIndustry,UniversityMalaysiaKelantan，KampusJeli,LockedBag,Jeli,KelantaMalaysia 2．SchoolofHealthSciences，HealthScience，UniversitySainsMalaysia，KubangKerian，Kelantan16150,Malaysia Received：February14，2013／Published：November20，2013 Abstract：Antioxidnatsplayna importantroleininhibitingnadscavengingfreeradicalsinhuman，providingprotectionagainstcellular damageinrelationtocnacerinitiation．Seaweedshavebeenprovedtohavehighantioxidantactivity．Thus，thisresearchwasca~iedout todeterminetheantioxidantnadanticancerpropertiesofedibleredseaweed，Gracilariamanilaensis(Gracilraiales，Rhodophyta)．The extractswerepreparedbySoxhletextractionusingorganicsolventswithdifierentpolarities．Theantioxidantactivitiesofextractswere determinedintemrsoftheirfreeradicalscavengingactiviytfRSAIC50)andtotalphenoliccontent(TPC)．Thecytotoxicactiviytof extractsweretestedagainsthumanovariancancercellline(Caov-3)，humanbreastcancercellline(MDA-MB-231andMCF·7)， humancervicalcellline(HeLa)，mousefibroblasteellline(L929)andMadin．Darbycaninekidney(MDCK)andthecellviabilityafter 72hincubationwasdeterminedbymethyleneblueassay．ThefindingsshowedthatacetoneextracthasthelowestDPPH ICsovalue followedbyethylacetateextract．BothextractsalsoshowedhighvaluesofTPC．Dichloromethaneextracthadthestrongestcytotoxm onMDA．MB．23l(53．90．g／mL4-5．59 mL)andHeLa(95．50gg／mL)．While，acetoneandethylacetateextractswerecytotoxicon MCF．7(66．07gg／mL)andCaov．3(69．67 mL-4-l3．94~tg／mL)．Itcouldbeconcludedthattheantioxidantandcytotoxicactivitiesof G．manilaensiswereinfluencedbytheytpesofsolventsusedandthushadapotentialtodevelopasacancerchemopreVentl
