Relationship between LymphVessels withTumor-associated Macrophages in Colorectal CarcinomaTissuesAbstractObjective:Distantmetastasisofcolorectalcarcinomaisasignificantfactorthatresult infailueroftreatmentanddeaththroughbloodandlymphaticmetastasis.Relationship betweenbloodvesselsandtumorshasbeenin-depthstudied,butforalongtime,lackof thehighsensitivityandspecificitymarkerthatidentifylymphaticmicrovesselleadedto researchesoflymphangiogenesislagbehindthatofbloodvesselangiogenesis.D2-40isa novelmonoclonalmouseanti-lymphaticendothelialmarkerantibldy.Itdoesn’tmarkthe bloodvesselendothelialcellandpossesseshighsensitivityandspecificity,soitmayusein thelymphvesselcounting.Macrophagethatinfiltrateintothetumorareknownas tumor-associatedmacrophage(TAM).Itplaysanimportantroleintumorgrowth,invasion, metastasisandrecidivism.Recently,researchesontherelationshipbetweenTAMand bloodvesselangiogenesisareextensive, which consider that TAMisabletopromoteblood vesselangiogenesis.However,researchesontherelationshipbetweenTAMand lymphangiogenesisareinsuffice,oneofpossiblereasonsislackofthespecificityantibldy thatmarklymphvessel.Thepurposeofthisstudywastoinvestigatetheexpressionof lymphvesselandtumor-associatedmacrophageinbenignandmalignanttissueof colorectaltumortoresearchtheirrelationshipandtrytoapproachmechanismof lymphangiogenesis.Methods:Fourty-eightspecimensresectedfrompatientswithbenignormalignant colorectaltumorinTheSecondAffiliatedHospitalofGuangZhouMedicalUniversity between1998and2000.Another13specimensofcolorectalnormaltissueregardasthe controlgroup.Allspecimenswereconfirmedbypathology.Theywereimmunohi- stochemicallystainedbyanti-lymphaticendothelialmarkerD2-40monoclonalantibodyandanti-macrophagemarkerCD68monoclonalantibody.Referedtothemethodof WeidnerandLeek,wecountedthenumberofD2-40andCD68.StatisticalanalyseswerecarriedoutusingSPSS11.5statisticalpackages.Countsdataareshownasmeans±SD.Differenceandcorrelationbetweentwogroupsweredeterminedbyt–testandspearman correlation .Statistical
