ABSTRACTARGONAUTE(AGO)proteinswerehighlyconservedandlargegenefamily.Asthecorecomponentsofthesilencingcomplex,theyareintegralplayersinallknownsmallRNA-directedregulatorypathways.Trans-actingsiRNA(ta-siRNA)areaclassofendogenoussiRNAsthatcantargetmRNAstocontrolplantgrowthanddevelopment.AG07iskeycomponentoftheTAS3ta-siRNApathway.AGO7actfunctioninplantbyta-siRNApathway.Itwasshowedthattheta-siRNApathwayplaysaconservedroleinthedeterminationoflateralorganpolarity.However,thecontributionoftheta-siRNApathwayincontrollingtomatoleafandfloraldevelopmentislargelyunknown.Therefore,CloningandfunctionalanalysisofAGO7homologueSlAGO7inSolanumlycopersicumwillfurtherilluminatetheroleofta-siRNApathwayintheregulationofplantgrowthanddevelopment.ToinvestigatethepotentialbiologicalfunctionofSlAGO7inplantdevelopment,thefollowingaspectshavebeencarriedoutinthisstudy:BasedonESTsequencesfromtomatogenomedatabase(SGN),thefull-lengthcDNAandgenomicDNAsequenceofSlAGO7wereisolatedbyusingRACEandPCR.ThebioinformaticsmethodswereusedtoanalyzethesequencecharacteristicsofSlAGO7.Theseresultsshowedthattheclonedfull-lengthcDNAofSlAGO7was3238bpinlength,containinga3003bpopenreadingframe(ORF)whichencodes1000aminoacids.Inaddition,thefulllengthofgenomicDNAcontainedthreeextronsandtwointrons.ThededucedSlAGO7proteinwithtwoconservedPAZandPIWIdomainshadthetypicalcharacteristicsofAGOfamily.ThisdeducedaminoacidhadahighidentitywiththeknownAGO7protein.PhylogeneticanalysisshowedthatSlAGO7wasclassifiedintotheZIPPY/AGO7subfamilyofAGOfamily.Real-timePCRmethodwasusedtoinvestigatethetemporalandspatialexpressionpatternofSlAGO7gene.TheseresultsindicatethattheexpressionoftheSlAGO7genewasdetectedinvegetativetissuessuchasrootsandstem,butnotinleaves.SlAGO7mainlyexpressedinflowerorgan(Budandflower),whileitsmRNAwasnotdetectedinfruit.Furthermore,SlAGO7wasfoundtobeexpressedinallthepartsofflowersatanthesisstages.TheexpressionlevelofSlAGO7increasesslightlyinsepalanddecreasesdramaticallyinstamen.ThesedatasuggestthatSlAGO7haspotentialfunctionto
