3．以NAF的现有合成方法为基础，尝试使用β-环糊精和柱层析硅胶作为催化剂，合成NAF及其两个光学异构体，解决原有手性源合成方法上存在的副反应很多和产物不容易分离的问题，并应用于之前设计的新型芳氧基哌嗪化合物对映异构体的合成。4．尝试设计一种合成2,3-二氢香豆素的串联反应，对其反应参数进行初步的优化。研究结果：1．合成了四组12个新型芳氧基哌嗪类化合物及其对映异构体，通过HPLC、ESI-MS、IR、1HNMR对所合成化合物进行了纯度和结构鉴定，并对其进行了绝对构型的测定，结果与设计的目标化合物结构一致。2．运用双报告基因荧光酶检测了12个新化合物的生物活性，结果表明这些目标化合物均有不同程度的α1-肾上腺素受体拮抗作用，其中几个化合物相对于对照药物哌唑嗪拮抗系数大，提示可能是一种潜在的α1-肾上腺素受体拮抗剂。分析数据发现，芳基哌嗪类化合物的手性中心对其活性有影响。对于合成的新的手性芳基哌嗪化合物，S构型的活性高于R构型，消旋体的活性介于两者之间。3．使用β-环糊精和柱层析硅胶作为催化剂，合成NAF及其两个对映异构体，收率和ee值都很高，并应用于设计的12个新化合物的合成，得到很好的效果。4．完成了一个合成有良好药物活性的3,4-二氢香豆素类衍生物的串联反应，优化反应条件，对产物进行了晶体结构解析。关键词：α1-AR拮抗剂；生物活性评价；NAF；β-环糊精；柱层析硅胶；双报告基因荧光素酶；串联反应ResearchbackgroundAbstractAsthepaceofaginginChina,benignprostatichyperplasia(BPH)isacommondiseaseagitatingmeninoldage.Accordingtodeepresearchinthisarea,ithadbeenfoundthatα1-ARisagoodtargetfordrugtreatment.Sothatα1-ARantagonisthasbecomeaveryeffectivedrugforBPH.Agroupofaryloxypoperazinederivativesisrepresentative.NAFisoneofaryloxypiperazinederivatives,whichwasmadebyBoehringerMannheimcompany.Itisanewtypeofα1-ARantagonistdrug.Ithasachiralcentersoithastwoenantiomers.Comparingwithprazosin,NAFnotonlyhasthesamecurativeeffect,butalsohassomedramaticcharacteristics:lighteruntowardreaction,longerpotency,conveniencesume,noimpactonweightwhileloweringbloodfat.Sothatitcanbeusedforthetreatmentofbenignprostatichyperplasia(BPH)andurinaryincontinence.Thispaperiswillingtodesignandsynthesizeanewgroupofaryloxypoperazinederivatives,basedonwellestablishedresearchinourlab,whichhaseffectionofα1-ARantagonist.Thendopreliminarybioactiveresearchforthesecompoundsandexplorenewsyntheticproceduresforthesecompounds.ResearchContentTodesignandsynthesize4group，totally12ofnewaryloxypoperazinederivatives,includingtheirenantiomers.TouseHPLC,ESI-MS,IR,1HNMRtoanalysetheirconstruction.TouseDual-luciferasereportergenedetectionmethodtodopreliminarybioactiveresearchforthese12compounds.BasedonthechiralpoolmethodofNAF,ItrybrandnewmethodsforsynthesizingNAFandnewlydesigned12compoundsbyusingβ-cyclodextrinandsilicagelasc
