HGF对大鼠肝部分切除后肝细胞和星状细胞作用 　　中图分类号：R332 文献标识码： A 文章编号： 1814-8824(2008)-9-0012-03 　　摘要:目的 探讨 HGF 对大鼠肝部分切除后促肝细胞再生与抑制肝星状细胞活化的作用与关系。方法 健康SD大鼠82只，随机分为正常对照组（A组）、肝部分切除组 (B组)、肝部分切除后HGF 干预组(C组)、肝部分切除后CCL4干预组(D组)、肝部分切除后CCL4与HGF共同干预组(E组)。除正常对照组外，其余各组均行肝大部分切除,切除量60 %。术后 HGF 干预组腹腔内注射HGF, CCL4干预组用 40% CCL4-色拉油皮下注射。 A组各取1/3分别于第1周、第2周和第8周取材； B组、C组各取1/2分别于第1周、第2周取材； D组、E组于第8周取材。心脏采血检测ALT、TP；取出肝脏称重；肝组织行HE 及Masson 染色，在光镜下观察病理及形态学变化，计数肝细胞核分裂指数，图像分析胶原纤维面积比； 采用免疫组化方法检测α-SMA的平均灰度值；在电镜下观察肝细胞、肝星状细胞超微结构变化。结论 大鼠肝部分切除后应用HGF可明显改善肝功能，促进肝再生。同时可抑制肝星状细胞活化，从而抑制肝纤维化的形成。 　　关键词 肝细胞生长因子 肝星状细胞 肝再生 　　 　　To Research the Effect of HGF on Faciliating Hepatocytes Growth and Inhibiting Hepatic Stellate Cells Activation After Partial Hepatectomy of Rats.Liu Bin ，Liu Heng-xing ，Wang Huan-zhen.An Yang Yin Du Xin Xing Hospital ， Anyang 455000， Henan ，China. 　　【Abstract】Objective To research the effects of HGF on facilitating hepatocytes growth and inhibiting hepatic stellate cells activation and their relation after Partial Hepatectomy of rats.Methods 82 rats were purchased from the Experimental Animals Center of Zheng Zhou University. They were randomly divided into five groups: normal control group (group A), partial hepatectomy group (group B)，group of HGF intervention after partial hepatectomy (group C), group of carbon tetrachloride intervention after hepatectomy (group D)，group of both HGF and carbon tetrachloride intervention after hepatectomy (group E). Except normal control group, all the other rats underwent operation of 60% partial hepatectomy. After operation, HGF were inject to the peritoneal cavity in group C and group E. Carbon tetrachloride-arachisoil solution at 40% concentration were given through hypodermic injection in group D and group E. Parts of rats in group A，B and C were executed after 1st week and 2ndweek, parts of rats in group A，D and E were executed after 8 weeks.
