人白介素24基因重组腺病毒载体构建与鉴定 　　【摘要】目的 构建含有人白介素24（hIL-24）基因的重组腺病毒载体，为下一步病理性瘢痕的基因治疗研究奠定实验基础。方法采用基因工程技术将hIL-24基因的cDNA亚克隆至穿梭质粒pAdTrack-CMV上,鉴定正确后在PAdEasy系统中进行细菌内同源重组，通过脂质体将正确重组体包裹并转染293A细胞以包装并扩增病毒。采用酶切及PCR方法对重组腺病毒进行鉴定。结果酶切和PCR结果证实hIL-24基因重组腺病毒载体构建成功并可在293A细胞中表达，病毒滴度达107pfu/ml。结论 成功构建了有较强感染能力的含hIL-24基因的重组腺病毒载体。 　　【关键词】hIL-24；腺病毒；病理性瘢痕 　　 　　The construction and Identification of Recombinant Adenovirus Vector of hIL-24LIANG Jie, HE Hai-tian PENG Zhi, et al. MethodsThe human IL-24 gene fragment was cloned into the shuttle plasmid pAdTrack-CMV to form the transfer vector by the method of homogenous recombination in bacteria of PAdEasy system. Then the right recombinant adenovirus was transfected into 293A cells using Lipofectine 2000. The recombinant adenovirus vector was identified by restriction enzyme digestion and polymerase chain reaction (PCR). ResultsRestriction endonuclease and PCR analysis confirmed that the human IL-24 gene was successfully inserted into the adenovirus vector. The titer of the recombinant adenovirus was 107pfu/ml. ConclusionThe recombinant adenovirus vector of hIL-24 was successfully constructed with highly infection. 　　【Key words】Human IL-24 gene; Adenovirus; Pathological scar 　　 　　人白介素24基因（hIL-24）是一种肿瘤抑制基因，通过多种途径抑制、诱导、细胞的生长，对肿瘤细胞有选择性杀伤作用而对正常组织几乎无毒副作用，并且具有调节免疫系统的功能，是当今人们研究的热点[1]。目前已经在多种肿瘤如前列腺癌、肺癌等的肿瘤基因治疗方面取得实验成功[2]。近年来许多研究表明病理性瘢痕的形成与细胞周期蛋白、细胞因子、癌基因等密切相关。然而hIL-24基因与病理性瘢痕的关系尚未见报道。我们过往的研究表明hIL-24基因在增生性瘢痕内的表达水平与正常皮肤组、普通瘢痕组的相比较明显降低，提示hIL-24基因在增生性瘢痕的形成过程中起着重要作用，可能与IL-24基因在组织中的表达低下有关[3]。本实验拟构建含有hIL-24基因的重组腺病毒载体，为下一步病理性瘢痕的基因治疗研究做基础。 　　 　　1材料与方法 　　 　　1.1 材料 　　1.1.1试剂：PmeI、PacI、BglⅡ及XhoI购自NEB公司；质粒提取试剂盒、胶回收试剂盒购自Omega公司；Lipofectamine 2000脂质体转染试剂盒购自Invitrogen公司；RT-PCR试剂盒及PCR产物回收试剂盒
