TET2蛋白下调5hmC在膀胱癌细胞T739中低表达 　　【摘要】 目的：研究膀胱尿路上皮癌细胞中TET2蛋白及5hmC的表达量相关作用。方法：采用Real-time PCR检测正常尿路上皮细胞和膀胱尿路上皮癌细胞中TET2 mRNA的表达，并同时采用细胞免疫化学染色检测正常尿路上皮细胞和膀胱尿路上皮癌细胞中5mC及5hmC表达，了解TET与膀胱癌的作用关系，并探讨其可能的作用机制。结果：TET2 mRNA在正常膀胱细胞T24中的表达（1.400 00±0.057 74，n=3），显著高于膀胱癌T739细胞[（0.866 70±0.120 20，n=3），P=0.016 1]；TET2蛋白在T739中表达（1.589 00±0.048 52，n=3）也明显低于T24[（1.310 00±0.049 33，n=3），P=0.015 7]。5-mC在两种细胞中的表达量比较差异无统计学意义（P=0.097 6）；5-hmC的表达量T739细胞明显低于T24细胞 中国论文网 /6/viewhtm　　[（1.840 00±0.055 68，n=3） VS （1.487 00±0.041 77，n=3），P=0.007 1）]。结论：TET2 mRNA和蛋白水平在膀胱肿瘤细胞中低表达，显著低于正常膀胱细胞；且TET2与5hmC低表达存在相关性。 　　【关键词】 TET2； 5hmC； 膀胱癌 　　TET2 Protein Downregulates Low Expression of 5hmC in Bladder Cancer Cell T739/XIE Meimao，WANG Zhongjun，HU Yingqiu，et al.//Medical Innovation of China，2018，15（08）：036-039 　　【Abstract】 Objective：To study the expression of TET2 protein and 5hmC in bladder urothelial carcinoma cells.Method：Real-time PCR was used to detect the expression of TET2 in normal urothelial cells and bladder urothelial carcinoma cells mRNA，and at the same time immunohistochemical staining was used to detect 5mC and 5hmC in normal urothelial cells and bladder urothelial carcinoma cells，TET and bladder cancer，and its possible mechanism of action was explored.Result：The expression of TET2 mRNA in normal bladder cells in 　　T24（1.400 00±0.057 74，n=3），was significantly higher than that of T739 cells of bladder cancer[（0.866 70 　　±0.120 20，n=3），P=0.016 1].The expression of TET2 protein in T739 was significantly lower than that of T24[（1.589 00±0.048 52，n=3） VS （1.310 00±0.049 33，n=3），P=0.015 7].There was no significant difference in the expression level of 5-mC between two kinds of cells（P=0.097 6）.The expression level of 5-hmC was significantly different，and the T739 cell group was significantly lower than that of T24 cell group[（1.840 00 　　±0.055 68，n=3） VS （1.487 00±0.041 77，n=3），P=0.007 1].Conclusion：TET2 mRNA and protein levels are low expression in bladder tumor cells，significantly lower than th
