EndosomalpH-sensitivePTXprodrugmicellarnanoparticles:designandapplicationAbstractEndosamalpH-sensitivePTXprodrugmicellarnanoparticles:designandapplicationAbstractInthisthesis,endosomalpH-activatablepaclitaxel(PTX)prodrugmicellarnanoparticlesweredesignedandpreparedbyconjugatingPTXontowatersolublepoly(ethyleneglycol)-b-poly(acrylicacid)(PEG-PAA)blockcopolymersviaanacid-labileacetalbondtoPAAblockandinvestigatedforpotentgrowthinhibitionofhumancancercellsinvitro.PTXwasreadilyconjugatedtoPEG-PAAwithhighdrugcontentsof21.6,27.0,and42.8wt.%(denotedasPTXprodrugs1,2and3,respectively)usingethylglycolvinylether(EGVE)asalinker.Theamphiphilicpolymercanself-assembledinphosphatebuffer(PB,pH7.4,10mM)intomono-dispersecore-shellmicellarnanoparticleswithaveragesizerangingfrom158.3to180.3nm,dependingonthecontentofPTX.Theseprodrugmicellarnanoparticleswerestableatphysiologicalconditionandavoidedtheburstreleaseofdrugefficiently.TheinvitroreleasestudiesshowedthatdrugreleasefromPTXprodrugnanoparticleswashighlypH-dependant,inwhichca.86.9%,66.4%and29.0%ofPTXwasreleasedfromPTXprodrug3at37?Cin48hatpH5.0,6.0,andpH7.4,respectively.MTTassaysshowedthatpH-sensitivePTXprodrug3nanoparticlesexhibitedhighanti-tumoreffecttoKBandHeLacells(halfmaximalinhibitoryconcentrations(IC50)of0.18and0.9?gPTXequiv./mL,respectively)aswellasPTX-resistantA549cells(A549/PTXR).Ascomparison,thevinylether-functionalizedPEG-PAAwasnon-toxicityevenwhentheconcentrationwasreachedto1.0mg/mL.Inthefollowing,usingthepresentPTXprodrugnanoparticlesasplatform,weinvestigatedthefolicaciddecoratedpH-sensitiveprodrugnanoparticlesbasedonprodrug3andfolate-PEG-b-poly(D,L-lactide)(FA-PEG-PLA),andphysicalloadingandsimultaneousreleaseofdoxorubicin(DOX).Theaveragesizewasof185~191nmfortheFA-decoratedprodrugnanoparticlesatFA-PEG-PLAcontentof20wt.%.Notably,theseprodrugnanoparticlesdisplayedapparenttargetabilitytofolatereceptor-overexpressingAbstractEndosomalpH-sensitivePTXprodrugmicellarnanoparticles:designandapplicationKBcells(-FA)withIC50over12timesl
