I I 摘 要 摘 要 本研究对替勃龙原料药的质量标准、替勃龙片剂的稳定性及生物样品中替勃龙含量 的检测方法进行了研究。 第一章：介绍了替勃龙的研究概况，对绝经期体内指标的变化及替勃龙对其治疗机 制进行了综述，介绍了替勃龙对绝经期临床症状的改善作用，并对其检测方法的国内外 研究进展做了综述，阐明了实验研究的目的与意义。 第二章：替勃龙原料药含量及有关物质检测方法的研究。 建立了反相高效液相色谱法（RP-HPLC）测定替勃龙原料药含量及有关物质的新方 法。考察了不同流动相对样品与有关物质分离度、灵敏度的影响，以及柱温、流速等对 检测结果的影响。结果表明，建立的 RP-HPLC 法，能够准确的测定原料药中样品和有 关物质的含量，本方法操作简便，灵敏度高、重现性好，结果准确可靠，可用于替勃龙 原料药含量以及有关物质的检测与分析，用于原料药的质量控制。 第三章：替勃龙片剂的稳定性考察及含量测定方法的研究。 采用经典恒温法，考察温度和 pH 值对替勃龙片剂稳定性的影响，并对替勃龙片剂 的含量进行了测定。该方法操作简便、快捷，重复性好，精密度高，测定结果准确、可 靠，可用于替勃龙片剂剂型的稳定性研究、含量测定及质量控制。 第四章：人血浆中替勃龙含量检测方法的研究。 采用反相高效液相色谱法检测人体血浆中替勃龙的含量。考察了样品预处理条件、 流动相组成、检测器参数等因素对检测结果的影响。结果表明，建立的 RP-HPLC 法， 能够使替勃龙在单一流动相、单流速、等度洗脱条件下取得良好的检测结果。实验采用 有效的样品预处理方法，大大提高了样品回收率。该方法简单、灵敏、准确，回收率高， 检出限低，分离度好，适合生物样品中替勃龙含量的检测分析。 关键词 RP-HPLC 替勃龙 原料药 有关物质 含量测定 替勃龙片 稳定性 人体血浆 II II Abstract Abstract In this paper, we established a method to determine the content of tibolone in biological sample, and explore the quality standard of tibolone bull drugs and the stability of tibolone tablets. Chapter One: This chapter describes the research situation of tibolone, the change of body index in postmenopausal women and the treatment mechanism of tibolone were reviewed. Introduced the effect of perimenopausal syndrome of tibolone, sums up the detection method’s researches both in inland and abroad, interprets the objective and significance of the experimental study. Chapter Two: The research of detection method about the content and its related substances of tibolone bull drug. It’s to established an reversed phase high performance liquid chromatography (RP-HPLC) method to determine the content and its related substances of tibolone. To investigate the effect of different mobile phase upon the separation and sensitivity, the effect of column temperature and velocity upon the final target detection result.
