加样时差对竞争抑制法检测抗HBc影响因素探讨 　　[摘要] 目的:探讨加样时差对ELISA一步法试剂检测抗-HBc结果的影响。方法:选择抗-HBc阴性的标本分为A、B、D 3组,选择抗-HBc阳性的标本设为C组。A组标本加入后放置室温不同时间再加入酶标试剂;B组标本加入后放置37℃水浴不同时间再加入酶标试剂;C组先加酶标试剂37℃水浴放置不同时间再加入标本;D组先将标本与酶标试剂混匀后加入放置室温不同时间。结果:A、B组加样方式对抗-HBc检测结果影响明显,且加标本与加酶标试剂的间隔时间越长,反应温度越高,标本的假阳性越多;C组加样方式对抗-HBc检测结果影响明显,且加酶标试剂与加标本的间隔时间越长,标本的假阴性越多;D组加样方式对抗-HBc检测结果无影响。结论:A、B、C 3组实验的加样时差导致的不公平竞争可对ELISA一步法检测试剂的结果产生影响,出现抗-HBc假阳性、假阴性增多;D组加样方式可最大限度地减少抗-HBc假阳性和假阴性,保证检验质量。 　　[关键词] 加样时差;酶联免疫吸附试验;乙型肝炎病毒核心抗体;假阳性 　　[中图分类号] R446.6 [文献标识码]B [文章编号]1674-4721(2010)04(a)-056-02 　　 　　Study the factors of the adds the tipe time difference to the competitive inhibition method for detection of anti-HBc 　　HE Ying,WU Renfeng 　　(Laboratory Medicine Hospital of Xuzhou, Jiangsu Province,Xuzhou 221008,China) 　　[Abstract] Objective: Discussed the influence adds the type time difference to exame the anti-HBc result to the ELISA one-stage process reagent.Methods:Chose anti-HBc the negative sign duty A,B,D three groups, chose the anti-HBc masculine specimen as C group.
