II II 首先,通过各实验组中正常小鼠餐后血糖的比较,发现紫甘薯花色苷高 (75mg/kg)、中(50mg/kg)剂量组与阿卡波糖阳性药组均可显著抑制正常小鼠餐 后血糖的升高且与空白对照组比较有显著性差异(P0.01),根据相关文献报道, 推断紫甘薯花色苷与阿卡波糖相似也是通过抑制体内 α-葡萄糖苷酶活性来达到降血 糖的功效。 采用四氧嘧啶造成高血糖小鼠模型,通过预实验确定高血糖小鼠的各实验组给 药剂量。给药 30 天后,通过测量各实验组(空白对照组,模型对照组,紫甘薯花色 苷高、中、低剂量组,格列本脲阳性组)小鼠空腹血糖,进行比较后发现紫甘薯高 剂量组(50mg/kg)具有显著降糖效果,与模型组相比有极显著差异(P0.01);中 剂量组( 25mg/kg )与模型组比较具有显著相差异( 0.01P0.05 );而小剂量 (12.5mg/kg)降糖效果不明显,与模型组无显著性差异。 对各组小鼠血清中的 SOD、MDA、GSH-Px 三个抗氧化指标的测定结果进行比 较发现,紫甘薯花色苷高剂量和中剂量组血清中的 MDA 含量比模型组显著下降 (P0.01),SOD、GSH-Px 含量比模型组显著上升(P0.01),而低剂量组的 SOD、 MDA、GSH-Px 与模型组相比也有显著性差异(0.01P0.05)通过对小鼠胰脏病理 切片的观察也发现,紫甘薯花色苷高、中、低剂量组都在一定程度上修复了小鼠胰 脏的腺泡细胞,并呈现一定的线性关系,随着紫甘薯花色苷剂量的增加,其修复效 果越好,这也进一步说明紫甘薯花色苷通过抗氧化功能修复被四氧嘧啶过氧化损伤 的腺泡细胞,增加胰岛素的分泌,达到降血糖的功效。 关键词:紫甘薯花色苷;双酶法;提取纯化工艺;降血糖功能及机理; III III Abstract With the widespread use of food coloring, more and more people further understanding the hazards of synthetic pigment, people began to pay close attention to the food coloring from natural sources. Because of its good coloring effects, as well as the different changing color, anthocyanin pigment has been widely used in food industry. In plants, anthocyanins often combined sugar through the glycosidic bond, existed in the form of anthocyanins. Anthocyanins not only can be used as a food coloring, because of its physiological effects, it also can be used as a food fortifier. Purple sweet potato, as a rich source of anthocyanins, has been one favorite food of more and more people in recent years. It has become a common food on people’s dining-table. In our study, the purple sweet potato as raw material, the extraction and purification of purple sweet potato anthocyanins, its hypoglycemic activity and mechanism are researched. First, determine the efficient, less pollution enzymatic extraction as the method of extracting purple sweet potato anthocyanins.
