成都中医药大学硕士学位论文 成都中医药大学硕士学位论文 PAGE PAGE 10 Abstract Study of blood stasis on the rat model of visceral hypersensitivity of irritable bowel syndrome Objective:Irritable bowel syndrome (IBS) is a common gastrointestinal diagnosis .The symptoms of IBS are similar to those of blood stasis. The purpose of this study was to investigate changes of hemorheology and endothelium function on the model of visceral hypersensitivity of IBS in rats, and to assess the relationship between blood stasis and visceral hypersensitivity of IBS. Methods: 30 neonatal female SD rats were randomly divided into three groups: model group, control group and Herba Leonuri group. The models and Herba Leonuri group received mechanical colonic stimulation between postnatal days 8 and 21.The threshold of abdominal withdrawal reflex (AWR) were recorded during colorectal distention at postnatal 35th day. Then Herba Leonuri group were treated by traditional Chinese herb Herba Leonuri(with dosage of 3.0 g/kg)for 4 weeks. Models and controls were given purified water respectively. The threshold of AWR were recorded again at postnatal 64th day. At the end of the experiment,hemorheology parameters and serum level of ET-1,NE and NO were examined. Result: 1.In contrast to the controls, model rats showed a significant decrease in the threshold of AWR both at postnatal 35th day and 64th day (P0.01); 2.The whole blood viscosity(in 1/200s、1/30s、1/5s、1/1s shear rate),the plasma viscosity, hematocrit, the whole blood relative viscosity(in high shear rate) and the plasma level of fibrinogen in model rats were obviously or significantly higher than that of controls (P0.05 or P0.01); PT and APTT significantly shorter(P0.01); 3.The serum levels of ET-1 and NE of models were significantly higher than that of control group(P0.01),and the serum levels of NO were significantly lower(P0.01); 4.Compared to model group, the threshold of AWR of Herba Leonuri group were significantly increased(P0.01);The whole blood vi
