法具体操作严格按照 ELISA 试剂操作说明进行。采用 winmdi 软件分析 SLE 患者 淋巴细胞上 P2X7R 的表达水平与对照组的差异；比较 SLE 患者不同临床特征组 P2X7R 的表达水平差异；分析P2X7R 与 IL-6 、TNF- α、IL-1 β的相关性。 结 果 SLE 患者外周血 CD4+、CD19+细胞表面 P2X7R 的表达明显高于健康对照 组,差异有统计学意义（CD4+细胞：2.21(3.55) vs 0.89(1.15) ，p =0.015 ；CD19+细胞： 11.53(20.01) vs 6.66(6.27)，p =0.037 ）；SLE 组血清中的细胞因子 IL-6、TNF- α及IL-1 β水平均明显高于对照组，差异有统计学意义；NPSLE 组血清中 IL-6 的水平较非 NPSLE 组明显升高，差异有统计学意义；28 例健康对照组外周血 P2X7R 的表达 与血清中 IL-6、TNF- α及IL-1 β的水平呈正比；29 例 SLE 患者外周血淋巴细胞中 P2X7R 表达与 IL-6 （r=0.449 ，p =0.015 ）呈正相关；初发 SLE 患者中，关节炎组 淋巴细胞上 P2X7R 表达显著高于非关节炎组(3.84 （11.53）vs0.90 （1.81）,p =0.006) ； 初发 SLE 患者中 P2X7R 在淋巴细胞及 CD19+细胞中的表达均与 SLEDAI 评分呈 正相关；淋巴细胞上的 P2X7R 表达与抗β2GP Ⅰ呈正相关（r=0.575 ，P=0.008 ）。 血清中 IL-6 的水平与 SLE 患者中尿蛋白的水平呈正相关（r=0.523 ，p =0.004 ）；血 清中 IL-6 及 IL-1 β的水平与SLEDAI 评分成正相关（分别为 r=0.435 ，p =0.021 ； r=0.418，p =0.027 ）。 结 论 P2X7R 可能通过介导炎症因子的释放参与 SLE 发病；可能与 SLE 患者的 关节炎、狼疮肾炎及神经精神狼疮相关。 关键词 嘌呤受体，离子通道，红斑狼疮，系统性，细胞因子 4 The expression of the P2X7 purinergic receptor on lymphocytes and its clinical significance in patients with new-oneset systemic lupus erythematosus Abstract Objective analyze the expression of the P2X7R on different lymphocyte subsets and its correlations with inflammatory cytokines in order to investigate their clinical significance in patients with SLE.
