应用盐溶蛋白电泳图谱鉴定玉米种子纯度-吉林农业大学学报.PDF

DO :10.13327/j .j j lau.2001.04.002 吉林农业大学学报　200 1, 23 (4):6 ～10 J ournal of J ilin Agr icu lt ur al Un iversi ty 方玉春, 张兴和, 冯德举, 刘俊平, 庄炳昌, 梁晓葵 (吉林省农业科学院玉米研究所, 吉林 公主岭 136100) 　:应用盐溶蛋白电泳方法对38 份玉米杂 种和37 份亲本自 系进行了蛋白电泳生化指纹图谱品种 真实性及其纯度检测。 结果表明:有34 份单 种能够很好地鉴定出来, 4 份单 种未能鉴定出来, 鉴定有效 率达89%;本方法还能鉴定亲本自 系的纯度及同一母本的不同单 种, 能有效监控假冒种子。 对凝胶谱带 胶板进行干燥处理, 从根本上解决了该项技术结果保存难的关键问题, 仅用3 ～ 4 d 就能准确建立品种的特征 谱带样板, 为该项技术的规范化应用和进一步研究奠定了基础。 该方法具有操作简便、快速、准确、经济、可批 量检测的特点, 适合在实际中应用。 :玉米;种子纯度;盐溶蛋白电泳;鉴定 :S513.037 　　　:A 　　　:1000-5684(200 1)04-0006-05 The Identificationof CornSeed Purity by Using PAGEPatterns of Salt-soluble Protein FANG Yu-chun, ZHANG Xing-he, FENG De-ju, L U Jun-ping, ZH UA NG Bing-chang, et al. (I nst it ute of Corn Research, J ilin A cademy of Agr icu ltura l S ciences, Gongz hu ling , J i lin 136100, Ch ina ) Abstract:T he truthf ulness and purity of 38 co rn hybrids and 37 corn inbred lines were analyzed by using PAGE patterns of salt-soluble protein and the biochemical f ingerprints of these varit ies w ere est ablished .T he result s show ed that:34 hybrids out of 38 could be distinguished effectively , namely , the effective rate of identification reached 89%.T his technology can also be used to de- termine the purity of inbred lines and is therefo re able t o provide effect ive basis for controlling fake seeds.The desiccative treat ment of glue boards of gel PAGE patterns tho roughly tackled the difficult problem of preserving t he results of this technology and it o nly took 3 ～4 days to estab- lish the standard patterns of variet ies.Such finding s can lay a foundation f or t he normalized uti- lization of this technology .Ow ing to its sim plicity in operation
