万方数据 万方数据 Abstract Objective: To locate and quantify gastric microbiota of patients with chronic gastritis and to explore the relationship between gastric mucosa microbiota with inflammation and Helicobacter pylori (Hp) infection. Methods: The genomic DNA was extracted from antral biopsies obtained from 58 patients with chronic gastritis. Total number of bacteria and Hp was quantified using real- time quantitative PCR. The gastric mucosa from additional 12 patients with chronic gastritis were embedded in paraffin, cut into slices to locate the gastric microbiota using Fluorescence in situ hybridization. The grading of chronic gastritis and intestinal metaplasia was based on the updated Sydney System. Results: Multiple linear regression analysis showed that the total number of bacteria was associated with Hp infection and the degree of chronic gastritis (P0.05), wheras it was irrelevant with sex, age and the grade of intestinal metaplasia. The real-time quantitative PCR showed the mean of Log value of the total number of bacteria per gram tissue was 10.00±0.42. It was significantly higher in Hp positive group (10.30±0.91) compared to negative group (9.64±0.78) (P0.01). Moreover, the total number of bacteria was positively correlated with the Hp quantity (r=0.536, P0.01) in Hp positive group. This suggested that Hp infection could influence gastric microbiota. The total bacteria number of the severe gastritis group was significantly higher than the mild and moderate (P0.05, P0.01), suggesting that gastric microbiota was likely to influence the inflammatory degree. Results of fluorescence in situ hybridization found that the gastric microbiota of patients with chronic gastritis mainly located on the surface of epithelial cells, gastric pits and in the glands. They distributed scatteredly or clamped together. Conclusions: Gastric microbita is associated with Hp infection and the grade of chronic gastritis, suggesting the increase number of gastric microbiota may involve
