Methods Total 76 patients pathological diagnosed with colorectal cancer by surgery were collected and the expression of Smad4 protein, TGF-βand TGF-βⅡR protein were measured in both specime of normal mucosa and tumor tissue with immunohistochemistry (IHC). The relatiohip between the expression of Smad4 protein, TGF-βand TGF-βⅡR protein and the clinic-pathological facto was analyzed. Resluts The positive rate of Smad4 protein expression in the tumor tissues and normal mucosa were % (27/76) and % (47/76) respectively and their difference was significant statistically (P). The positive rate of TGF-βprotein in tumor tissue and normal mucosa were % (19/76) and % (32/76) respectively and their difference is significant statistically too (P). The positive rate of TGF-βⅡR protein in tumor and normal mucosa were % (14/76) and % (16/76) respectively, there was no significant difference statistically (P); The high level expression of Smad4 protein in tumor tissue was relative to the earlier tumor stage and fewer lymph node metastasis（P）. There was no relatiohip between Smad4 protein and gender, age, tumor location, tumor size, differentiation, tumor embolus, pathologic type, as well as TGF-βⅡR、TGF-βprotein (P). Conclusion Smad4 and TGF-βprotein are down-expressed in tumor tissue in contrast to normal mucosa in colorectal cancer patients and Smad4 proteins lower expression is always accompanied with later stage, more lymph node metastasis, But there is no significant differe
