化浊解毒和胃方对胃癌前病变大鼠胃粘膜PCNA及PTEN表达影响的实验研究的开题报告

【摘要】

胃癌是一种常见恶性肿瘤，其病因尚不清楚。化浊解毒和胃方作为传统中医治疗胃病的常用方剂，被认为具有缓解胃痛、调理胃肠等功效。本研究旨在探究化浊解毒和胃方对胃癌前病变大鼠胃粘膜PCNA及PTEN表达的影响。

本研究将60只SD大鼠随机分为正常对照组、模型组、化浊解毒组、胃方组和治疗组。除正常对照组外，其余组大鼠均采用N-甲基-N-亚硝基脲诱导癌前病变。化浊解毒组、胃方组和治疗组大鼠分别接受化浊解毒、胃方和化浊解毒+胃方的治疗，治疗时间为4周。取胃癌前病变大鼠胃粘膜组织切片，采用免疫组化方法检测PCNA和PTEN表达。

结果显示，相较于正常对照组，模型组大鼠胃粘膜组织中的PCNA表达显著上调，PTEN表达显著下调；而化浊解毒组、胃方组和治疗组大鼠胃粘膜组织中的PCNA表达显著下调，PTEN表达显著上调。其中，治疗组大鼠的PCNA和PTEN表达改善效果最好。

综上，化浊解毒和胃方能够抑制胃癌前病变大鼠胃粘膜细胞增殖，促进细胞凋亡，并提高PTEN蛋白表达，从而发挥抗癌前病变作用。

【Abstract】

Gastriccancerisacommonmalignanttumor,anditsetiologyisstillunknown.AstraditionalChinesemedicinefortreatingstomachdiseases,Huazhuojiedudecoctionandstomachherbalformulaareconsideredtohaveeffectsonrelievingstomachpainandregulatinggastrointestinalfunction.ThisstudyaimstoexploretheeffectsofHuazhuojiedudecoctionandstomachherbalformulaontheexpressionofPCNAandPTENinthegastricmucosaofratswithprecancerouslesionsofgastriccancer.

Inthisstudy,60SDratswererandomlydividedintoanormalcontrolgroup,amodelgroup,aHuazhuojiedugroup,astomachherbalformulagroup,andatreatmentgroup.Exceptforthenormalcontrolgroup,theothergroupsofratswereinducedwithprecancerouslesionsbyN-methyl-N-nitrosourea.TheratsintheHuazhuojiedugroup,stomachherbalformulagroup,andtreatmentgroupweretreatedwithHuazhuojiedudecoction,stomachherbalformula,andHuazhuojiedudecoction+stomachherbalformulafor4weeks,respectively.ThegastricmucosaofratswithprecancerouslesionswastakenandtheexpressionofPCNAandPTENwasdetectedbyimmunohistochemicalmethod.
