【Abstract】AIM:TodeterminethepossibleeffectsofdifferentdosesofinsulinontheproinflammatorycytokineIL1βandtheantiinflammatorycytokineIL10inearlystageofsevereburninrats.METHODS:MaleSpragueDawlyrats(30%TBSA,fullthicknessburn)wererandomlydividedinto2groupstoreceiveeithernormalsalineonly(n=14,withintraperitonealinjectionof40mL/kgnormalsaline,groupA)ornormalsalineplusdifferentdosesofinsulin(n=42,withintraperitonealinjectionof40mL/kgnormalsalineplussubcutaneousinjectionof1,3or5IU/kginsulinrespectively,groupB1,B2,B3).Inaddition,7ratswithoutthermalinjuryweresubjectedtonormalcontrolgroup(groupN).The
outcomemeasuresencompassedtheserumglucose,bloodpressure,serumIL1βandIL10inratsat6and24hafterscaldanddifferenttreatments.RESULTS:Theserumglucoseincreasedquicklyinratsafterscald,whichwasdemonstratedingroupAtoarangeof(10.7±1.8)mmol/Lby6hpostscald.InsulindecreasedtheserumglucoseinadosedependentmannerinratsofgroupB1,B2andB3.Andinsulinatadoseof3IU/kgbodyweightreducedtheserumglucosetoarangeof(6.0±0.8)mmol/Linratsat6hafterthermalinjury,whichwasalmosteqealtothenormalrangeof(5.1±0.6)mmol/L(Pgt;0.05).Meanwhile,insulinatthisdoseimprovedthebloodpressureofscaldedratsbykeepingthebloodpressureatastableconditionsimilartothenormal.
InsulinsignificantlydecreasedtheserumIL1βinadosedependentmannerinscaldedrats.Itwaspartiallyshowedafteradministrationofinsulinof3IU/kgserumIL1βwas(54.0±8.6)ng/Land(46.8±6.8)ng/L,muchlowerthanthatwithoutinsulinadminist
