Abstract：Effectofstrophanthidin (Str)onintracellularcalcium concentration ([ca ]；)was
investigatedon isolatedventricularmyocytesofguineapig．Singleventricularmyocyteswereobtainedby
enzymaticdissociationtechnique．Fluorescentsignalof[ca ]．wasdetectedwithconfocalmicroscopy
afterincubationofcardiomycytesinTyrode’ssolutionwithFluo3-AM．TheresultshowedthatStrincreased
[ca ]iinaconcentration-dependentmanner．Theventricularmyocytesbegantoround-upintoa
contracturestateoncethepeaklevelof[ca ]iwasachievedinthepresenceofStr(10I~mol·L )，but
remainednochangeinthepresenceofStr(1and100nmol·L )．Tetrodotoxin(TTX)，nisodipine，
andhighconcentrationofextracellularCa“changedtheresponseofcardiomycytestoStr (1and100
nmol·L一)，buthadnoobviouseffectsontheactionofStr(10I~mol·L一)．Theelevationof[ca ]i
causedbyStratallofthedetectedconcentrationswaspartiallyantagonizedbyrynodine(10I~mol·L )
orthe removalofCa from Tyrode’ssolution． In Na ， K -free Tyrode’ssolution，theresponse of
cardiomycytesin[ca ]；elevationtoStr(10I~mol·L )wasattenuated，whileremainednochangeto
Str(1and100nmol·L )．TTX，nisodipine，andhighconcentrationofextracellularCa“changedthe
responseofcardiomycytestoStratallofthedetectedconcentrationsinNa ，K -freeTyrode’ssolution．
Thestudysuggeststhattheelevationof[ca ]；byStratthelow (nomomolar)concentrationsispartially
mediatedbytheextracellularcalcium influxthroughCa“channelora “slip modeconductance’’ofTTX
sensitiveNa channe1．Whiletheeffecto
