大肠腺癌中基质金属蛋白酶2、9及其抑制剂1、2的表达 作者：陈谦，柯娟，王纪全，邓怀慈 【摘要】 目的 探讨人大肠腺癌组织中基质金属蛋白酶2、9(MMP2、9)及其抑制剂1、2(TIMP1、2)的表达及其临床意义。方法 采用免疫组织化学SP法检测MMP2、MMP9和TIMP1、TIMP2在30例大肠腺癌组织及相应切缘正常组织中的蛋白表达。结果 大肠腺癌组织中MMP2、MMP9及TIMP1的阳性表达明显高于正常切缘组织(Plt;0.05)，并与肿瘤分化程度、浸润深度、Dukes分期以及淋巴结转移呈正相关(Plt;0.05);TIMP2在大肠腺癌组织中低表达(Plt;0.05)，并与浸润深度、Dukes分期以及淋巴结转移呈负相关(Plt;0.05)，与肿瘤分化程度无明显相关性(Pgt;0.05);MMP9和TIMP1表达明显相关(r=0.4924, Plt;0.05)。结论 MMP2、MMP9及TIMP1的过度表达与大肠腺癌的侵袭、转移有密切关系，其联合检测有助于评估大肠腺癌的恶性生物学行为及预后。 【关键词】 大肠癌;基质金属蛋白酶;组织金属蛋白酶抑制剂;免疫组织化学 　　ABSTRACT: Objective To study the expressions of matrix metalloproteinase2 (MMP2), matrix metalloproteinase9 (MMP9) and their inhibitors, tissue inhibitor of metalloproteinase TIMP1 and TIMP2, in human colorectal cancer and their clinical significance. Methods The expressions of MMP2, MMP9, TIMP1 and TIMP2 in 30 primary colorectal tumors and adjacent noncancerous tissues were detected by immunohistochemical SP method. Results The positive rates of MMP2, MMP9 and TIMP1 in colorectal cancer were significantly higher than those in adjacent noncancerous tissues (Plt;0.05), and were closely associated with tumor differentiation, Dukes stages, progression of invasion depth and lymph node metastasis (Plt;0.05). The positive rate of TIMP2 was much lower in colorectal tissues than in adjacent noncancerous tissues (Plt;0.05), and was negatively correlated with Dukes stages, progression of invasion depth and lymph node metastasis (Plt;0.05), but not to tumor differentiation (Pgt;0.05). A close correlation was present between MMP9 and TIMP1 expression (r=0.4924, Plt;0.05). Conclusion Overexpressions of MMP2, MMP9 and TIMP1 are obviously correlated with both invasion and metastasis of colorectal cancer, combined detection of which may be helpful in determining the biological behavior and prognosis of colorectal cancer. 　　KEY WORDS: colorectal cancer; matrix metalloproteinase; tissue inhibitor of metalloproteinase; immunoh
