临床医学论文-甲状腺癌中端粒酶逆转录酶mRNA和Survivin的表达及意义 ????????????????? 作者：钱婀娜，徐正顺，王仰坤?? 【摘要】? 　　目的： 探讨甲状腺癌（TC）中端粒酶逆转录酶（TERT）mRNA和survivin蛋白的表达与细胞增殖和凋亡的关系. 方法：采用原位杂交法、末端脱氧核苷酸酶标记法及免疫组化技术检测46例TC中TERT mRNA、细胞凋亡、增殖细胞核抗原（PCNA）和survivin蛋白的表达，20例良性病变作为对照. 结果： TC的TERT mRNA和survivin的表达阳性率,增殖指数（PI）,凋亡指数（AI）均显著高于对照组；TC中TERT mRNA和survivin的表达均与临床分期有关，survivin还与淋巴结转移有关；二者的表达呈显著正相关；TERT mRNA和survivin阳性表达的TC组织AI明显低于阴性组，而PI均显著高于阴性组（P＜0.05）. 结论： TERT和survivin特异性的表达于TC组织中，可能通过促细胞增殖和抗凋亡途径，参与TC的发生发展. 【关键词】? 甲状腺肿瘤 细胞凋亡 细胞增殖 端粒酶逆转录酶 存活素蛋白 　　【Abstract】 AIM: To investigate the expressions of survivin protein and telomerase reverse transcriptase (TERT) mRNA and their relationships with cellular proliferation and apoptosis in thyroid carcinoma(TC). METHODS: The terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end labeling technique was introduced to measure the apoptosis index(AI). Expressions of TERT mRNA and the protein of survivin,proliferating cell nuclear antigen (PCNA) in 46 cases of thyroid carcinoma were respectively determined by in situ hybridization and immunohistochemical staining. Twenty cases of benign lesions were taken as control group. RESULTS：The positive staining rates of TERT mRNA,survivin and AI,proliferation index(PI) in TC were higher than those in control group (P＜0.05).The expressions of TERT mRNA and survivin in TC were related to clinic stage(P＜0.05). At the same time,the expression of survivin was related to lymph node metastasis. In TC group,the expression of TERT mRNA was positively correlated with the expression of survivin. In the TC group,PI in patients with positive survivin or TERT mRNA expression was higher than that in patients with negative survivin or TERT mRNA expression,however,the AI was lower than that in patients with negative survivin or TERT mRNA expression（P＜0.05）.
