陈年普洱茶特征成分的生物活性研究

摘 要:采用顶空-固相微萃取-气质联用法测定陈年普洱茶中挥发性组分,结合质谱和保留指数共鉴定出58种化合物,其以甲氧基酚类、醇类及酮类化合物为主。采用比色法及高效液相色谱测得猛景、康藏和宝焰牌普洱沱茶的总多酚、可溶性总糖、游离氨基酸、儿茶素类、生物碱及没食子酸的含量。清除DPPH自由基实验表明三种普洱茶均具有较好的清除DPPH自由基能力,成分分析说明高浓度的普洱茶提取物中起主要清除作用的成分为没食子酸。MTT结果显示三种普洱茶提取物对人肝癌细胞SMMC-7721的增值均有明显的抑制作用,且呈剂量依赖性,其中宝焰牌普洱沱茶抑制癌细胞生长效果最为明显,提取物浓度为250μg/ml时,对人肝癌细胞的抑制作用高于90%。

Analysis on Characteristic Flavor Components and Bio-activity of Aged Pu-erh Tea

Abstract: Volatile compound of aged Pu-erh tea was analyzed by solid-phase micro-extraction (SPME) combined with gas chromatography-mass spectrometry (GC-MS). Results showed that a total of 58 volatile compounds were identified by matching EI mass spectra and retention indices with the literature data. Alcohols, methoxy-phenolic and ketones were three major aromatic groups characterized. Contents of total polyphenols, soluble saccharide, free amino acid, gallic acid, catechins and polyphenols in Pu-erh tea samples were analyzed. All types Pu-erh tea extracts showed exhibited the relative strong DPPH· scavenging activity. The components analysis demonstrated that gallic acid is the major component responsible for scavenging DPPH free radical from high concentrations of pu-erh tea extracts. Results from the MTT assay revealed that all the three kinds of Pu-erh tea extracts significantly inhibited the proliferation of SMMC-7721 cells,in dosage-dependent modes. More than 90% inhibitory effect of 250μg/ml baoyan pu-erh tea extract on the proliferation of SMMC-7721 cells were observed in this study.
