MiR-122 sequestration by its antagomir leads to decreased cholesterol levels, low density lipoprotein (LDL) and high density lipoprotein (HDL) fractions, and triglyceride content in the both liver and blood. In addition, miR-122 silencing or the functional inhibition of miR-122 by anti-miR- 122 oligonucleotides causes potent and sustained inhibition of HCV replication. On the contrary, over-expression and restoration of miR-122 can stimulate the replication of HCV, but may inhibit the replication of HBV and the growth and metastasis of HCC, also sensitize HCC cells to chemotherapeutic agents. It’s worthy to note that miR-122 deficiency may result in steatohepatitis and liver fibrosis. Therefore, miR-122-based therapeutics might be a double-edged sword to the liver diseases due to its contradictory roles in different liver diseases.
