Shh和Ptch在结肠癌、腺瘤性息肉中的表达及意义 　　作者：张　平，喻文人　 作者单位：(重庆市荣昌县人民医院消化内科，重庆　402460) 　　【摘要】 目的：检测Shh和Ptch蛋白在结肠癌、腺瘤性息肉中的表达，探讨二者在结肠癌发生、发展的作用，关注其在不同临床病理特征中的表达差别，为临床工作提供理论支持。方法：以80例大肠癌、80例结肠腺瘤性息肉、80例正常结肠黏膜组织作为研究对象，采用免疫组织化学技术，检测Shh和Ptch蛋白的表达，比较二者在不同临床指标及临床病理特征之间的关系。结果：Shh和Ptch蛋白在正常结肠黏膜组织、腺瘤性息肉、结肠癌中高的表达依次增高，Shh和Ptch在结肠癌中的表达与其肿瘤的有无炎细胞浸润、分化程度、Dukes分期及淋巴结转移密切相关，结肠癌中Shh和Ptch的表达呈正相关。结论：Shh和Ptch蛋白在结肠癌中高表达，显示二者在结肠癌的发生、发展中起协同作用，联合检测结肠癌中Shh和Ptch蛋白的表达可能与患者的预后相关。 　　【关键词】 结肠癌;Shh;Ptch;免疫组织化学 　　[ABSTRACT] Objective： To explore the expression difference of Shh and Ptch protein in colorectal carcinoma and neoplasm， as well as their roles in the development and progress of colorectal carcinoma. Methods： The study included involved tissues collected from 80 cases of colorectal carcinoma (observation group)， 80 cases of adenomatous polyp (control group)， as well as 80 cases of normal colorectal mucosa (normal control group). Expressions of Shh and Ptch in the three groups were detected and observed by IHC. Clinical parameters and pathological characters were observed and compared among these groups. Results： The expressions of Shh and Ptch were the highest in colorectal carcinoma， followed by adenomatous polyp and normal colorectal mucosa. The expressions of Shh and Ptch were correlated with inflammatory cell infiltration， differentiation， dukes stage and lymph node metastasis. Positive correlation between Shh and Ptch were found in colorectal carcinoma. Conclusions： The higher expressions of Shh and Ptch may promote the occurrence and development of colorectal carcinoma. The combined examination may be helpful to predict the prognosis in colorectal carcinoma. 　　[KEY WORDS] Colorectal carcinoma; Shh; Ptch; Immunohistochemistry (IHC) 　　结肠癌临床常见，是严重危害人类健康的肿瘤，但是其发生机制仍不明确近年来人们对其发病机制进行了深入研究，最初学者发现Hedgehog(Hh)信号通路在动物胚胎发育过程中调节细胞间的相互作用[1,2]，但随着研究的深入，其不仅仅在胚胎发育过程中有作用，同时其信号通路在多种肿瘤中的表达呈显著激活状态[3,4]。本实验应用免疫组织化学技术检测80例结肠癌组织中Shh和Ptch蛋白的表达情况，关注二者在肿瘤中的表达特点及关系，旨在为研究肿瘤的发生、发展提供理论帮助。 　　1　材料和方法
