2012年 2月第 20卷第 2期 MODERN0NC0L0GY．Feb．2012．VOI．20．N0．02 · 217 · 囹论著囹 ◆：．基础研究．．．不同来源骨髓间质干细胞分离培养及成骨分化比较赵志正，林洪生 Isolation，cultureandosteogenicdifferentiationofdifferentsourcesofmesenchymalstemcells ZHAO Zhizheng，LIN Hongsheng OntologyDepartment，GuanganmenHospital，ChinaAcademyofChineseMedicalSciences，Being100053，China． A【bstract】 Objective：Mesenchymalstemcells(MSCs)aremultipotentadultstemceils．Inrecentyears，more andmoreconcernshavebeendrawnonMSCsinthecancer—relatedfield．Soweisolated，identifiedandpurifeddif- ferentsourcesofbonemarrowmesenchymalstem cells．comparetheirosteogenicdifferentiationofpluripotentdifferen· tiationdifferences，chooseagoodcellsourceforthenextstudy．M ethods：UsingdifferentialadhesionseparationSD ratsandBalb／emicebonemarrow mesenchymalstem cells，purifiedbypassage，andobservethepurificationand proliferationofMSCsthoughtheprocessofstatus．IdentifytwokindsofMDSCscellsurfaceantigenexpressionbyflow cytometry．Comparetheabilityofosteogenicdifferentiationbetweendifferentsourcesofbonemarrow mesenchymal stem cells．Results：SD ratsduringisolationandcultureofMDSCs，theproliferationwassignificantlystrongerthan Balb／emiceMDSCs，thetypicalswirlingpostedwallgrowthcharacteristicscanbeseeninthethirdgeneration．By flowcytometry，Balb／cmiceandSD ratsbothexpressMesenchymalstem ceilsmarker．InadipogenieSD ratsMDSCs stillshowed astrongerability，SD ratsMDSCshadsignificantlyearliertimeofshowingmineralizationnodesthan Balb／emiceMDSCs，andSD ratsMDSCsexhibitedagreaterextentthanBalb／cmiceMDSCs．Conclusion：SDrats wereeasiertoobtainhighpurityandproliferativecellofMDSCs．TheSDratsMDSCshavestrongeradipogeniccapaci— tythantheBalb／cmiceMDSCs，toacertainextent，itcanbeagoodsourceofcellsofrstudy． K【eywords】mesenchymMstemcells(MSCs)；cellisolationandculture；identification；osteogenicdifferentiation
