学院代号 10716 学 号 专业代码 1002 密 级 Shaanxi University of Chinese Medicine 学士学位论文 Box-Behnken效应面法结合HPLC指纹 图谱优选酒制太白山茱萸炮制工艺 学位申请人 专业名称 中药制药 申请学位类型 学士学位 指导教师姓名 论文提交日期 2008年7月 中文摘要 目的:研究太白山茱萸炮制工艺参数,设计响应面,并以马钱苷、莫诺干含量,结合指纹图谱优化其蒸制工艺条件。方法:采用高效液相色谱法,测定以山茱萸不同的蒸制工艺参数所得样品的图谱中马钱苷含量,莫诺苷含量结合指纹图谱为指标,通过3因素3水平的Box-Behnken响应面试验设计,考察加酒量、闷润时间、蒸制时间等因素对其炮制效果的影响;色谱柱 Diamonsil C18 (250 mm×4.6 mm,5 μm),柱温30 ℃;流动相 A:乙腈 B:0.1 %磷酸水溶液,梯度洗脱;流速为1.0 mL/min,Waters2996二极管阵列检测器,检测波长240 nm。结果:以全自动高效液相色谱法测定17批不同蒸制工艺的山茱萸药材谱图,经方法学考察和响应面设计,结果r=0.9998,莫诺苷在0.714~1.666 mg/mL的范围内吸收度与浓度呈现良好的线性关系, r=0.9994;马钱苷在0.0672~0.448 mg/mL的范围内吸收度与浓度呈现良好的线性关系,考察指标成分为马钱苷含量,莫诺苷含量,结合指纹图谱综合评定蒸制工艺。结论:本方法简洁、准确,易操作,酒蒸制山茱萸较清蒸工艺所得制品更优,且确定最佳加酒量为20 %,闷润时间为2 h,蒸制时间为6 h。 关键词 莫诺苷;马钱苷;指纹图谱;Box-Behnken效应面法。 Abstract Objective: To study the Taibai Mountain of cornel processing parameters, and loganin, Monod dry content, combined with the fingerprint of the steaming process condition optimization.
