The effect of chalcones extracted from Angelica Keiskei on the mRNA
expression of insulin signal key molecules in hepatocytes of rats with
够pe 2 diabetes
ABSTRACT
Objective:To observe the effect of chalcones extracted from Angelica Keiskei(AC)on
the mRNA expression of insulin receptor(InsR)、insulin receptor substrate一2(IRS-2)、
Phosphatidy I inositol 3-kinase(P13K)and serine-threonine(Akt)and explore Ashitaba
chalcone improvement in type 2 diabetic rats insulin signal transduction disorder.
Method:The diabetes of rats was induced by streptozotocin with intraperitoeal injection
as well as with high—fat diet feeding.The rats were randomly divided into 3 groups诚th
1 0 rats in each group,diabetic control group,high-dose AC group and low-dose AC
group.All the rats were fed、Ⅳith high—fat diet.0.30 and 1 0 mg/kg。BW AC per day were
given to high-dose AC,low—dose AC and the diabetic control groups,respectively.Another
1 0 normal rats fed with regular diet Was used as the normal control group.After 4 weeks,
serum insulin levels were evaluated by radioimmunoassay.The mRNA expression levels
of InsR、IRS一2,P13K and Aktwere determined by reverse transcription polymerase
chain reaction(RT-PCR).The protein expression levels of insulin receptor in hepatocytes
were detected by immunohistochemistry.and the phosphorylation levels of Akt in
hepmocytes were detected by western blot.
Results:
(1)the levels of fasting blood glucose and fasting serum insulin:the level of fasting
blood glucose in rats ofhigh-dose AC gruop Was 7.0 4-1.12,insulin level Was 29.5 4-5.3l,
compared with the diabetic control group,the difference Was statistically significant(P
0。05).
(2)insulin resistance index:the high and low dose groups insulin resistance index
were:8.99 4-O.79,23.47 4-5.65,diabetic control group was 28.06 4-3.24,the difference
between groups was statistically significant(F=1 3,26,PO.05).High dose groups were
compared with the diabetic control group,the difference was statistically significant(P
