Tianetal.VirologyJournal2011,8:121 /content/8/1/121 RESEARCH OpenAccess Comparativeanalysisofoncogenicgenesrevealed uniqueevolutionaryfeaturesoffieldMarek’s diseasevirusprevalentinrecentyearsinChina MingxingTian ,YangZhao,YanLin,NianliZou,ChengLiu ,PingLiu ,SanjieCao ,XintianWen1,2, 1 1 1 1 1 1,2 1,2 YongHuang1,2* Abstract Background:Marek’sdisease(MD)isaneconomicallyimportantviraldiseaseofchickenscausedbyMarek’s diseasevirus(MDV),anoncogenicherpesvirus.Thisdiseasewaswellcontrolledsincethewidespreaduseof commercialvaccines,butfieldMDVshaveshowncontinuousincreasinginvirulenceandacquiredtheabilityto overcometheimmuneresponseinducedbyvaccines.Nowadays,MDcontinuestobeaseriousthreattopoultry industry,isolationandcharacterizationofMDVsareessentialformonitoringchangesofvirusesandevaluatingthe effectivenessofexistingvaccines. Results:Between2008and2010,18fieldMDVstrainswereisolatedfromvaccinatedchickenflocksinSichuan province,China.ThreeoncogenicgenesincludingMeq,pp38andvIL-8genesofthe18isolateswereamplified andsequenced.Homologyanalysisshowedthatthededucedaminoacidsequencesofthesethreegenesexhibit 95.0-98.8%,99.3-100%and97.0-98.5%homologyrespectivelywiththeseofotherreferencestrainspublishedin GenBank.Alignmentanalysisofthenucleotideanddeducedaminoacidsequencesshowedthatfouraminoacid mutationsinMeqgeneandtwoaminoacidmutationsinvIL-8genedisplayedperfectregularityinMDVs circulatinginChina,whichcouldbeconsideredasfeaturesoffieldMDVsprevalentinrecentyearsinChina.In addition,oneaminoacidmutationinpp38genecanbeconsideredasafeatureofvirulentMDVsfromUSA,and threeaminoacidmutationsinMeqgenewereidentifiedanduniqueinveryvirulentplus(vv+)MDVs. PhylogeneticanalysisbasedonMeqandvIL-8proteinsequencesrevealedthatfieldMDVsinChinaevolved independently.VirulencestudiesshowedthatCVI988couldprovideefficientprotectionagainstthefieldMDVs epidemicrecentlyinChina. Conclusions:ThisstudyandotherpublisheddataintheGenBankhavedemonstratedthefeaturesofMeq,pp38 andvIL-8genesofMDVscirculatinginr
