UDK619:575.224 CYTOGENETICANALYSISOFTHEEFFECTSOFEPINEPHRINEONCULTUREDHUMAN LYMPHOCYTES DJELI]N*,DJELI]DIJANA*,SPREMO-POTPAREVI]BILJANA**,MARKOVI]BILJANA* and @IVKOVI]LADA** *FacultyofVeterinaryMedicine,UniversityofBelgrade, **FacultyofPharmacy,UniversityofBelgrade (Received2.February,2003) Epinephrine can modulate mitotic activity of normal and malig- nantcellsandexhibitgenotoxicpotentialinsometest-systems.Itisas- sumedthatmetabolicconversionofphenolicgroupsinthecatechol ringofepinephrineleadstotheformationofreactivederivativesandsu- peroxide anions, capable of damaging cellular molecules including DNA. Theaimofthepresentstudywastoevaluatethecytotoxicand genotoxiceffectsofepinephrineonhumanperipheralbloodlympho- cytesinvitro.Thelowestconcentrationofepinephrineusedintheseex- -10 periments(5x10 M)wascalculatedtobeintherangeofthephysio- logicalbloodlevelofepinephrineinhumans.Threeexperimentalcon- -7 -6 centrationscorrespondedtominimal(2x10 M),average(10 M)and -6 maximal(5x10 M)therapeuticdosesinhumanmedicine.Inaddition, thehighestconcentrationsexceededthemaximaltherapeuticdose10- -5 -4 fold(5x10 M)and30-fold(1.5x10 M),respectively.Onthebasisof theresultsobtaineditcanbeconcludedthatepinephrinehadnoinflu- ence on the appearance of chromosome aberrations under the de- scribed experimental conditions. However, mitotic index was signifi- cantlylowerinculturestreatedwiththethreehighestconcentrationsof epinephrineusedinthisinvestigation.
