Andrographolide(AND), thediterpene lactonecompound, waspuri?ed by HPLCfromthe methanolicfractionof theplant Andrographis paniculata. The compound was found to have potent antiplasmodial activity when tested in isolation and in combinationwithcurcuminandartesunateagainsttheerythrocyticstagesofPlasmodiumfalciparuminvitroandPlasmodium berghei ANKA in vivo. IC50s for artesunate (AS), andrographolide (AND), and curcumin (CUR) were found to be 0.05, 9.1 and17.4μM,respectively.Thecompound(AND)wasfoundsynergisticwithcurcumin(CUR)andaddictivelyinteractivewith artesunate(AS).Invivo,andrographolide-curcuminexhibitedbetterantimalarialactivity,notonlybyreducingparasitemia(29%), comparedtothecontrol(81%),butalsobyextendingthelifespanby2-3folds.Beingnontoxictotheinvivosystemthisagentcan beusedastemplatemoleculefordesigningnewderivativeswithimprovedantimalarialproperties. 1.Introduction and anticancer properties [12].
